New NO‐Donors with Antithrombotic and Vasodilating Activities, Part 17. Arylazoamidoximes and 3‐Arylazo‐1,2,4‐oxadiazol‐5‐ones

Abstract: Seven arylazoamidoximes (3), six phenoxycarbonyl derivatives (4), and six 1,2,4-oxadiazol-5-ones (5) have been prepared and their structure and purity established by spectroscopy and elemental analysis. In the EI mass spectra ready elimination of NO from the title amidoximes was observed. A new addition reaction of 3a with hydrochloric acid to 4-chlorophenylhydro-azoamidoxime 7 is described. The compounds were tested for nitric oxide dependent biological properties, i.e. platelet aggregation, antithrombotic ef… Show more

“…An arylazo group directly linked to the amidoxime group yielded compounds with considerable antithrombotic and antihypertensive activities via NO generation [132]. The same team has extended the study to amidoximes with other electron withdrawing substituents [133] as well, and the results in terms of structure-activity relationships are given below.…”

“…The oxidative formation of nitric oxide from amidoximes by liver cells [132] was mimicked in vitro by the addition of a Fe 3+ -porphyrin complex (as cytochrome surrogate) and an oxygen donor, i.e. (bis-trifluoroacetoxy)-iodo-benzene to the ethanolic solution of selected compounds.…”

“…Due to their NO-donating capacities, some oxime derivatives have also been shown to offer therapeutic potential for the treatment of erectile dysfunction, as well as cardiovascular diseases [ 203 , 204 ]. Likewise, a number of oxime derivatives have been shown to exhibit antithrombogenic, hypotensive, and cardiotonic activity [ 198 , 205 , 206 ]. For example, amidoximes and oximes have been shown to inhibit platelet aggregation, decrease thrombus formation, induce vasodilation, and lower intraocular pressure [ 192 , 197 , 199 , 207 , 208 , 209 ].…”

“…The first major works that appeared on that topic were published by Rehse et al in 1997 and 1998 [66,67]. Seven aryl azoamidoximes and their effects on thrombosis inhibition, blood pressure and inhibition of platelet aggregation were investigated [66]. Five of the seven amidoximes inhibited the formation of thrombus in mesenteric rat vessels and even two of these amidoximes inhibited the thrombus formation by more than 20% (Figure 10).…”
