New Multicomponent Crystal Forms of Adiphenine with Low Hygroscopicity

Abstract: Adiphenine is an acetylcholine receptor inhibitor used as an antispasmodic drug due to its strong smooth muscle relaxant action. Adiphenine hydrochloride is largely marketed in drug associations to treat muscle spasms and relieve pain in colic and cramps. However, it presents serious problems with hygroscopicity and chemical stability under high humidity conditions, which have limited its use as an active pharmaceutical ingredient (API). Here, we have solved the adiphenine solid-state hygroscopicity problem th… Show more

“…A similar observation was noted with the palmatine saccharinate salt and gallic acid-palmatine chloride co-crystal, which showed better hygroscopic stability but reduced its aqueous solubility [ 23 , 35 ]. Another study by Ribeiro et al [ 15 ] aimed at reducing the hygroscopicity of adiphenine hydrochloride by preparing more stable oxalate and citrate salt forms. This greatly enhanced the stability of the molecule but the aqueous solubility reduced significantly.…”

“…Various control strategies have been employed to improve the hygroscopic stability of susceptible APIs ( Figure 1 ), such as the use of manufacturing controls [ 11 ], protective packaging of dosage forms [ 12 ], coating with hydrophobic polymers/lipids [ 13 ], encapsulation in lipid formulations [ 14 ], changing salt forms [ 15 ], formation of co-crystals [ 16 ], etc. The advantages and disadvantages of each control strategy are outlined in Table 1 .…”
