New Knowledge About CCR5, HIV Infection, and Disease Progression: Is “Old” Still Valuable?

Abstract: CC chemokine receptor (CCR) 5 (CCR5) is the main HIV-1 coreceptor involved in virus entry and cell-to-cell spread during acute and chronic infections: such CCR5 and T cell tropic viruses are adapted to and replicate in CD4 + memory T cells. Polymorphisms in CCR5 regulate CCR5 expression, which, in turn, influences HIV infection acquisition and subsequent disease progression. Among these polymorphisms, a 32-bp deletion in the CCR5 open reading frame (CCR5 D32) and a single nucleotide polymorphism (SNP) in the p… Show more

“…CCR5 D32 is a common mutation in CCR5 that can lead to a change in protein function and the loss of HIV co-receptor function. Several studies have shown that the CCR5D32 gene locus can effectively control infection (99)(100)(101)(102). In addition, some research results (103) indicate that the expression of CXCR2, CXCR4, and CCR5 are significantly changed in the peripheral blood mDC and pDC of HIV+ progressors.…”

“…Our findings also indicated no significant difference in IFN-γ expression level between the two groups of patients. In line with this finding, it has been shown that IFN-γ has no direct antiviral activity against HIV-1 infection in primary cultures, as supported by the in vivo findings of IFN-γ therapy in infected subjects [ 10 , 23 ] . In contrast, certain studies have reported that this cytokine can increase the cytotoxic T lymphocytes and natural killer cell activities against HIV-1-infected cells.…”

“…The major co-receptor -CCR5-provides natural resistance to HIV infection with 32bp deletion in both alleles (1,2). Recent studies showed gene modification and cell engineering of CCR5 can also induce resistance to HIV infection (10,(13)(14)(15)(16). Finding a suitable donor with HLA-matched by Δ32 target genotype is one of the challenges in allogeneic transplantation (17).…”
