New insights into TNFα/PTP1B and PPARγ pathway through RNF213- a link between inflammation, obesity, insulin resistance and Moyamoya disease

Abstract: AbstractDiabetic patients are always at a higher risk of ischemic diseases like coronary artery diseases. One such ischemic carotid artery disease is Moyamoya. Moyamoya disease (MMD) has been associated with diabetes Type-I and II and the causality was unclear. RNF213 is the major susceptible gene for MMD. To understand the association between diabetes mellitus and MMD we chose the major players from both the anomalies, insulin and RNF213. But before establishing a role of RNF2… Show more

“…Baseline expression of RNF213 is observed in many tissues [18] but expression is upregulated by LPS, TNFα, and interferon (IFN), in line with the idea that inflammatory and immune signals might act as environmental triggers for MMD [62][63][64][65]. RNF213 is present in the cytosol [18], but overexpression, oleic acid treatment [57], IFN, or LPS stimulation [56,64] relocate the protein to the surface of LDs, organelles that mediate lipid storage but also function as intracellular innate immune hubs [64].…”

“…Upstream, RNF213 is negatively regulated by protein tyrosine phosphatase 1B (PTP1B or PTPN1), and this was found to be important for hypoxia sensing and the control of nonmitochondrial oxygen consumption in tumors [69]. Also in adipocytes, RNF213 is regulated by PTP1B following TNFα stimulation, potentially mediated by peroxisome proliferator-activated receptor gamma (PPARγ) [65].…”

“…These findings suggest that transcriptional regulation of RNF213 seems to be cell type-and context-dependent. In macrophage or adipocyte, another inflammatory component, lipopolysaccharide (LPS), was shown to upregulate RNF213, while peroxisome proliferatoractivated receptor γ (PPARγ), an anti-inflammatory signal, downregulated it (53). In adipocyte, PTP1B mediated higher expression of RNF213 by TNF-α, while PTP1B suppresses E3 ligase activity of RNF213 in a breast cancer cell line under hypoxic condition (54) (Supplementary Figure 1).…”
