New Insights for Diagnosis of Pineapple Fusariosis by MALDI-TOF MS Technique

Abstract: Fusarium is one of the most economically important fungal genus, since it includes many pathogenic species which cause a wide range of plant diseases. Morphological or molecular biology identification of Fusarium species is a limiting step in the fast diagnosis and treatment of plant disease caused by these fungi. Mass spectrometry by matrix-assisted laser/desorption ionisation-time-of-flight (MALDI-TOF)-based fingerprinting approach was applied to the fungal growth monitoring and direct detection of strain Fu… Show more

“…Additionally, mass-spectrometry-based approaches have also been applied successfully for the identification and differentiation of Fusarium species based on spore protein profiles [ 37 , 38 ]. Furthermore, MALDI-TOF MS peptide/protein fingerprints have been used to distinguish closely related species in the Fusarium fujikuroi species complex [ 39 ], and for in situ diagnosis in infected plant tissue [ 40 ]. Here, we have tested the potential of MALDI-TOF MS to distinguish the used Fusarium species based on protein fingerprints of mycelium.…”

“…Fusariosis caused by Fusarium guttiforme (formerly Fusarium moniliforme var. subglutinans, Fusarium subglutinans) appeared in Brazil in the 1960s and became the major concern for producers in that region [9][10][11][12][13]. The name "Fusarium guttiforme" brought about confusion, and some articles dealing with FCR used it to describe the pathogen [14,15].…”

“…The polyphasic approach combines the use of different techniques, such as morphology, biochemical and molecular biology tools and more recently, mass spectrometry (particularly matrix-assisted laser desorption/ionisation time-of-flight mass spectrometry (MALDI-TOF MS). Furthermore, mass spectrometry based microbial spectral analysis has been used as a key step in the polyphasic identification of filamentous fungi from different fields [12,14,22,23,24,25,26,27,28].…”
