New horizons in life extension, healthspan extension and exceptional longevity

Abstract: Many common chronic diseases and syndromes are ageing-related. This raises the prospect that therapeutic agents that target the biological changes of ageing will prevent or delay multiple diseases with a single therapy. Gerotherapeutic drugs are those that target pathways involved in ageing, with the aims of reducing the burden of ageing-related diseases and increasing lifespan and healthspan. The approach to discovering gerotherapeutic drugs is similar to that used to discover drugs for diseases. This include… Show more

“…Despite this, research into agerelated diseases aimed at slowing, halting and even reversing the aging process has not stopped, and extending the healthy lifespan of humans remains the focus of anti-aging research. 28,29 Researchers have been trying to find out which molecules and mechanisms are involved in the ageing process so that more corresponding drugs and sensible dietary supplements can be produced. Senescence of senescent cells or telomere attrition is an important cause of senescence, and therefore, anti-aging strategies by scavenging senescent cells and reducing the attrition of telomeres are also potential pharmacological strategies.…”

“…For this study, the primary challenge facing to us is how to measure the accelerated aging effect. The most common clinical outcomes in current anti-aging clinical studies mainly include age-related diseases and aging-related biomarkers (54,55). Distinguishing from chronological age, biological age as a biomarker of aging is a promising surrogate endpoint for anti-aging therapies in clinical research, such as KDM BioAge and PhenoAge (56,57).…”

“…As we are seeing with research on the biology of ageing (geroscience 2 ), age-related chronic diseases are not inevitable or untreatable; there are common pathways, defined by the hallmarks of ageing, 3 including cell senescence and systemic chronic inflammation, that lead to multiple morbidities. These pathways can now be manipulated with drugs and other interventions 4 early in the disease process, with promising data emerging from early human clinical trials. 5 We need to take a multipronged whole-of-life approach to tackle all the different, but interacting, drivers of health and disease.…”
