New horizons for in vitro spermatogenesis? An update on novel three-dimensional culture systems as tools for meiotic and post-meiotic differentiation of testicular germ cells

Abstract: Culture and differentiation of male germ cells has been performed for various purposes in the past. To date, none of the studies aimed at in vitro spermatogenesis has resulted in a sufficient number of mature gametes. Numerous studies have revealed worthy pieces of information, building up a body of information on conditions that are required to maintain and mature male germ cells in vitro. In this review, we report on previously published and unpublished experiments addressing murine germ cell differentiation… Show more

“…The reconstitution of spermatogenesis in vitro from SSC suspensions has proven more challenging. Some groups have obtained sperm in vitro by using testicular cell suspensions from prepubertal animals but three dimensional support matrices are necessary [35,36] . Although these successful systems have been developed for sperm production .…”

“…Likewise, there were other studies using 3D cell culture system to gain more insights into the interactions among germ cells, somatic cells and various extracellular matrices for the in vitro spermatogenesis (Hue et al 1998, Tanaka et al 2003, Stukenborg et al 2009, Abu Elhija et al 2012. The 3D blood-testis barrier model was also used for rat germ cell differentiation with production of haploid cells (Legendre et al 2010).…”

“…The production of mature eggs (mouse) from very small follicles [135] and fish (eel) spermatozoa from spermatogonia [136] is possible today. According to Stukenborg et al [137], mouse spermatogenic cells before entering meiosis can develop in vitro to morphologically normal spermatozoa. Perhaps within several decades we should be able to mass produce fully mature eggs and spermatozoa of economically valuable animals any time of the year.…”
