New Cocrystal of Ubiquinol with High Stability to Oxidation

Abstract: A new cocrystal of the essential nutrient ubiquinol that is highly resistant to air oxidation has been designed and discovered in a cocrystal screening. Hydroxybenzoic acids and phenols have been selected as the coformers with the aim to transfer their molecular antioxidant properties to the cocrystal and protect ubiquinol from oxidation. The new cocrystal ubiquinol/3,4-dihydroxybenzoic acid shows a remarkably high stability under standard stress conditions, which offers alternative strategies of formulation f… Show more

“…It is critical to develop an effective strategy to eliminate or minimize drug degradants. Recently, pharmaceutical cocrystals have emerged as a prospective approach to overcome the chemical instability of APIs in the solid state 100 , 101 . Table 1 shows the list of cocrystals with enhanced photostability.…”

“…That is, a significant change in the physicochemical properties of the API can be accomplished without its covalent modification. This technique has been used to improve pharmaceutically important properties of pharmaceutical and nutraceutical compounds in terms of hygroscopicity, bioavailability [2], stability [3,4], mechanical properties, etc. [5][6][7].…”

“…One pharmaceutical technology for enhancing the bioavailability is cocrystallization [ 26 , 27 ], which has also been used to improve drug physicochemical properties, such as solubility [ 28 , 29 , 30 ], dissolution rate [ 31 , 32 , 33 ], stability [ 34 ] and tabletability [ 35 , 36 ]. MTZ cocrystals with pyrogallol [ 37 ], 2,5-dihydroxy-benzoic acid, 2,6-dihydroxy-benzoic acid and 3,5-dihydroxy-benzoic acid [ 38 ], and gallic acid [ 3 ] have been reported.…”
