New Bioactive Heteroleptic Copper(II) Carboxylates: Structure, Enzymatic and DNA-Binding Studies

Abstract: Two new binuclear O-bridged copper(II) carboxylates with chemical formulas [Cu 2 (3-ClC 6 H 4 CH 2 COO) 4 (phen) 2 ] (1) and [Cu 2 (3-ClC 6 H 4 CH 2 COO) 4 (bipy) 2 ] (2) where phen = 1,10-phenanthroline and bipy = 2,2'-bipyridine have been synthesized and characterized by FT-IR, UV-Visible spectroscopy, CHN analysis and single crystal XRD. The results revealed distorted square pyramidal geometry around each copper atom of 1 and 2. The DNA interaction studies showed strong binding with K b = 5.07 × 10 3 and 4.… Show more

“…The ratio of slope to intercept of the plot of the terms on the left‐hand side of Equation (3) against 1/[DNA] (shown in Figure A) gave the K b value equal to 1.384 × 10 4 M –1 . This value has been found comparable to those for other dinuclear copper(II) complexes and indicates potent DNA‐binding ability of the complex.…”

“…24 Cu-O distances of the complex are quite asymmetrical. The Cu-O bond distances in the equatorial plane are 1.9441(16) and 1.9800(15) Å which are typical of the Cu-O equatorial distances of the previously reported dimeric 25 and polymeric 26,27 complexes. The axial Cu-O distance is 2.3535(15) Å indicating relatively weaker bond with copper.…”

“…This Cu-O-Cu asymmetric bonding of the bridging oxygen is also typical of other copper(II) complexes. 25 The smallest angle around Cu is N-Cu-N = 80.74(7) о being formed by the copper ion and two nitrogen atoms of bipyridine molecule and has been found typical of other copper(II) complexes with N-and O-donor ligands. 30…”

“…UV–visible absorption spectroscopy is the simplest and most commonly employed instrumental technique for studying both the stability of DNA and their interactions with small ligand molecules. Unlike large biomolecules which interact with DNA through binding with protein or RNA; small aromatic ligand molecules non‐covalently interact with DNA double helical structures by (i) intercalating between stacked base pairs thereby distorting the DNA backbone conformation or (ii) as minor groove binders which cause little distortion of the DNA backbone …”
