New Application of the Commercially Available Dye Celestine Blue B as a Sensitive and Selective Fluorescent “Turn-On” Probe for Endogenous Detection of HOCl and Reactive Halogenated Species

Abstract: Hypochlorous acid (HOCl) derived from hydrogen peroxide and chloride anion by myeloperoxidase (MPO) plays a significant role in physiological and pathological processes. Herein we report a phenoxazine-based fluorescent probe Celestine Blue B (CB) that is applicable for HOCl detection in living cells and for assaying the chlorinating activity of MPO. A remarkable selectivity and sensitivity (limit of detection is 32 nM), along with a rapid “turn-on” response of CB to HOCl was demonstrated. Furthermore, the prob… Show more

“…MPO chlorinating activity was evaluated fluorometrically using Celestine Blue B (CB) which was oxidized to fluorescent glycol (λ ex = 487 nm, λ em = 578 nm) by taurine N-chloramine produced in the reaction of taurine with HOCl generated by MPO [ 39 ]. The assay was performed in 96-plate format.…”

“…Celestine blue dye is used with iron-aluminium complexes as a substitute for haematoxylin in H-E (haematoxylin-eosin) staining because of its resistance to low-pH solutions. It has been used as a new electroactive indicator in DNA biosensors and is also applicable to HOCl detection in living cells and to assaying the chlorinating activity of myeloperoxidase [40]. In microbiology, neutral red is used in the MacConkey agar to differentiate bacteria for lactose fermentation [36].…”
