Neutrophil chemotactic activity elaborated by human dental plaque

Abstract: In the present investigation an in vivo method is described for studying the chemotactic activity elaborated by factors in human dental plaque. Plaque was sampled from the buccal and lingual surfaces of teeth from dental students, who had refrained from mechanical tooth cleansing during one 4 and one 8 day period. The plaque samples were suspended in 0.15 M NaCl, homogenized, and centrifuged at 12,100 × g for 30 minutes at 4°C. The plaque supernatant was then separated from the cellular pellet and sterile filt… Show more

“…5) the Limulus test gave no reaction ( Table 3). The samples (10)(11)(12)(13)(14)(15) pl) with gram-negative organisms could be diluted 104-106 times and still give a positive reaction. The more gramnegative cells in the sample, the higher degree of dilution could be obtained.…”

“…Wound chambers -Four perforated sterile cylinders of Teflon® (volume of 800 |il) were implanted subcutaneously in the back of each of 20 albino rats, as described by LINDHE & HELLDEN (1972). After 7 d, when the wound chambers had been surrounded by a granulation tissue of rich vascularity, the exudate of each chamber was aspirated and the volume determined.…”

“…Plaque was sampled after 7 and 14 d of abstaining from oral hygiene. The chemotactic activity for rabbit neutrophil leukocytes of the plaque formed during the two test periods was determined using an in vitro system described by BOYDEN (1962) and in accordance with a technique presented by LiNDHE & HELLDEN (1972).…”
