Neurotoxic effects of domoic acid on dopaminergic neurons in primary mesencephalic cell culture

Abstract: Introduction: Domoic acid is a potent marine neurotoxin produced by certain species of the diatom genus Pseudonitzschia. To our knowledge, there are no studies that have investigated neurotoxic effects of domoic acid on dopaminergic neurons. Accordingly, the present study was carried out to investigate the potential neurotoxic effects of domoic acid on dopaminergic neurons in primary mesencephalic cell culture. Material and methods: Cultures prepared from embryonic mouse mesencephala (total of 250 embryos) wer… Show more

“…Unlike MPP + which specifically damages dopaminergic neurons, rotenone was shown to injure other neuronal populations in primary mesencephalic cell culture. Similar results were obtained by our laboratory for some other neurotoxins like domoic acid [56] and acrylamide [57].…”

“…On gestation day 14, pregnant mice were killed, the uteri were dissected and the embryos were carefully removed to a Petri dish containing Dulbecco's phosphate buffered saline (DPBS, Invitrogen, Germany, pH 7.2). Brains were then obtained, mesencephala were excised and cultures were prepared according to Radad et al [20]. In brief, mesencephala were cleaned from meninges and cut into small pieces in a drop of DPBS.…”

“…DA can not only induce persistent epilepsy in rats but also promote aggressive behavior in rats [54]. Significant destruction of dopaminergic neurons is shown after long-term treatment of primary mesencephalic cell culture with DA [55]. Besides its toxicity on the nervous system, DA may also exert toxicological effects on other organs since most of the human organs have some level of glutamate receptor expression [56].…”
