NeuroSAFE PROOF: study protocol for a single-blinded, IDEAL stage 3, multi-centre, randomised controlled trial of NeuroSAFE robotic-assisted radical prostatectomy versus standard robotic-assisted radical prostatectomy in men with localized prostate cancer

Abstract: Background Robotic radical prostatectomy (RARP) is a first-line curative treatment option for localized prostate cancer. Postoperative erectile dysfunction and urinary incontinence are common associated adverse side effects that can negatively impact patients’ quality of life. Preserving the lateral neurovascular bundles (NS) during RARP improves functional outcomes. However, selecting men for NS may be difficult when there is concern about incurring in positive surgical margin (PSM) which in t… Show more

“…Another trial still in progress is the NeuroSAFE PROOF trial, a multicentre randomized controlled trial that aims to look into the effect of NeuroSAFE technique on functional outcomes, including incontinence and erectile dysfunction. Aiming to recruit 404 patients, the results are still pending [23]. The adoption of NeuroSAFE technique could assist in intraoperative decision-making and hence better functional outcomes for RARP for high risk or locally advanced cases.…”

“…However, it remains uncertain whether this translates to better functional outcomes. The NeuroSAFE PROOF trial (https://www.neurosafeproof.com/) will generate Level 1 evidence to answer this question [3]. Recruitment is complete, and the study is in the follow‐up phase with results expected in mid‐2024.…”

“…To evaluate the accuracy of LaserSAFE, we processed RARP specimens from patients enrolled in the control arm of the NeuroSAFE PROOF (NCT03317990) study [3] who signed an optional informed consent form, from April 2022 to February 2023. All patients underwent RARP using the DaVinci X Surgical System® (Intuitive Surgical Inc., Sunnyvale, CA, USA).…”
