Neuroprotective potential of isothiocyanates in an in vitro model of neuroinflammation

Abstract: Isothiocyanates (ITCs), present as glucosinolate precursors in cruciferous vegetables, have shown anti-inflammatory, antioxidant and anticarcinogenic activities. Here, we compared the effects of three different ITCs on ROS production and on the expression of matrix metalloproteinase (MMP)-2 and -9, which represent important pathogenetic factors of various neurological diseases. Primary cultures of rat astrocytes were activated by LPS and simultaneously treated with different doses of Allyl isothiocyanate (AITC… Show more

“…Also, AITC inhibited the activation of GFAP responsible for astrogliosis as well as preserved BBB integrity [ 107 ]. In a study that compared the effects of SFN, PEITC, and AITC on neuroinflammation, all ITCs were found to decrease the expression of MMP-2 and MMP-9, though only SFN significantly reduced circulating MMP-2 level via downregulation of ERK phosphorylation [ 291 ]. Interestingly, the effects of AITC upon binding to bitter taste receptor (TAS2R) were shown to be individual specific.…”

“…Benzyl isothiocyanate and 6-(Methylsulfinyl)hexyl isothiocyanate derived from Wasabi, have been reported to suppress a transcriptional levels of COX-2, an enzyme synthesizing the pro-inflammatory mediators ( Lee et al, 2009 ; Uto et al, 2012 ). The disturbance of phosphorylated MAPKs signaling, ERK, p38 kinase, and JNK, was observed in the ITCs treatment, resulting in the downregulation of the transcription of inflammatory genes such as COX-2, iNOS, TNF-α, IL-1β, and IL-6 ( Lee et al, 2009 ; Latronico et al, 2021 ). The effects of allyl-isothiocyanate and SFN on the Nrf2 nuclear translocation were associated with the downregulation of p65 protein, a subunit of the transcription factor NF-κB ( Wagner et al, 2012 ).…”

“…Detection of intracellular free radicals was performed by loading PBMC (2 × 10 5 cells/well in 96-well plates) or monocytes (2 × 10 5 cells/well in 96-well plates) with 10 µM 2',7'-dichlorofluorescein diacetate in phenol redfree IMDM at 37°C for 30 min (Latronico et al, 2021), then treated for 90 min with ConA (at final concentration of 5 µg/mL), LPS (at final concentration of 10 µg/ mL), or H 2 O 2 (at a final concentration of 1 mM). After incubation, the culture medium was removed and cells were rinsed twice with PBS.…”
