Neuroprotective Effects of the Sonic Hedgehog Signaling Pathway in Ischemic Injury through Promotion of Synaptic and Neuronal Health

Abstract: Cerebral ischemia is a common cerebrovascular condition which often induces neuronal apoptosis, leading to brain damage. The sonic hedgehog (Shh) signaling pathway has been reported to be involved in ischemic stroke, but the underlying mechanisms have not been fully elucidated. In the present study, we demonstrated that expressions of Shh, Ptch, and Gli-1 were significantly downregulated at 24 h following oxygen-glucose deprivation (OGD) injury in neurons in vitro, effects which were associated with increasing… Show more

“…On the contrary, the bottom genes had five significant enriched terms; TGF-beta signaling pathway, homologous recombination, globo sphingolipid metabolism, intraflagellar transport proteins binding to dynein, and DNA replication. Overall, the top twenty MPxgb(AD) seem relevant in neurological pathways 71 . In contrast, the bottom twenty are relevant in metabolism and DNA mechanisms; this does not omit neurological processes (Supplementary Fig.…”

“…Smo agonists have been demonstrated to play intricate roles in regenerative therapies [ 37 , 55 ] and purmorphamine has been shown to provide neuroprotective effects and greatly influence recovery of cognitive performance in rodents following neurological insult and aged postoperative cognitive dysfunction [ 22 , 37 , 56 ]. We examined the regulation of TBI-induced excitotoxicity through the mechanistic link between Shh signaling and Eaat2a.…”

“…Following injury, components of the Shh signaling pathway were rapidly and highly upregulated, and Shh was found to be important in the injury-induced proliferation. The use of purmorphamine, a Smoothened agonist and Shh signaling activator, was previously shown to ameliorate a variety of disorders, improve behavioral and cognitive outcomes, reduce neuroinflammation, and minimize edema following CNS trauma [ 22 , 23 ]. However, few studies have examined the effects of modulating Shh signaling in relation to PTS and regulating excitotoxicity.…”

“…The effect of this compound has been widely investigated in different models. It has been reported as resveratrol administration in rats, and exerts a protective effect against stroke by activating the neuroprotective pathway mediated by sonic hedgehog (Shh) [83,84]. In particular, 7 days of resveratrol pretreatment before MCAO significantly reduces infarct size and improves neurological function in rats [83].…”
