Neuroprotection of ischemic preconditioning is mediated by thioredoxin 2 in the hippocampal CA1 region following a subsequent transient cerebral ischemia

Abstract: Preconditioning by brief ischemic episode induces tolerance to a subsequent lethal ischemic insult, and it has been suggested that reactive oxygen species are involved in this phenomenon. Thioredoxin 2 (Trx2), a small protein with redox-regulating function, shows cytoprotective roles against oxidative stress. Here, we had focused on the role of Trx2 in ischemic preconditioning (IPC)-mediated neuroprotection against oxidative stress followed by a subsequent lethal transient cerebral ischemia. Animals used in th… Show more

“…To quantitatively analyze neuronal death/loss or protection, five sections of the anterior horn of the lumbar spinal cord were selected with 120 µm intervals in each animal. CV-and FJB-positive neurons were counted as previously described [24]. Briefly, digital images of the cells were obtained using a BX53 upright microscope (Olympus, Japan) equipped with digital camera (DP7) (Olympus, Japan) connected to PC monitor.…”

“…Limb ischemic preconditioning alleviates ischemia reperfusion injury, improves tolerance of ischemia, and delays or reduces subsequent longer ischemia reperfusion injury. Limb ischemic preconditioning can promote the release of a variety of endogenous active substances, such as adenosine, NO, endorphin, inhibiting neutrophil chemotaxis [ 22 ], and superoxide anion production [ 23 ].…”

“…DHE (Sigma-Aldrich, St. Louis, MO, USA) is the most popular fluorogenic probe to detect intracellular superoxide anions and is used to analyze oxidative stress. We applied DHE staining in the brain sections, as described previously [47]. Briefly, the sections were equilibrated in a Krebs-HEPES buffer (composed of 130 mM NaCl, 5.6 mM KCl, 2 mM CaCl 2 , 0.24 mM MgCl 2 , 8.3 mM HEPES, 11 mM glucose, pH 7.4, etc.)…”
