Neuronal Adenosine A1 Receptor is Critical for Olfactory Function but Unable to Attenuate Olfactory Dysfunction in Neuroinflammation

Abstract: Adenine nucleotides, such as adenosine triphosphate (ATP), adenosine diphosphate (ADP), as well as the nucleoside adenosine are important modulators of neuronal function by engaging P1 and P2 purinergic receptors. In mitral cells, signaling of the G protein-coupled P1 receptor adenosine 1 receptor (A1R) affects the olfactory sensory pathway by regulating high voltage-activated calcium channels and two-pore domain potassium (K2P) channels. The inflammation of the central nervous system (CNS) impairs the olfacto… Show more

“…Mice were acclimatized two weeks before EAE induction in the experimental housing facility. EAE induction was performed as described before (31). Briefly, 13 to 14-week-old female C57Bl/6J mice were anesthetized with isoflurane (1% to 2% v/v in oxygen).…”

“…Both A 1 and A 2A receptors are the main adenosine receptors expressed in the olfactory bulb and modulating neuronal performance (31,37,38). Olfactory bulb astrocytes express A 2A receptors that mediate an increase in the cytosolic Ca 2+ concentration (39).…”
