Neuron-Like Differentiation of PC12 Cells Treated With Media Conditioned by Either Sciatic Nerves, Optic Nerves, or Schwann Cells

Abstract: In previous works we reported the finding of neurotrophic activity in a serum-free Dulbecco's modified Eagle's medium conditioned by rat sciatic nerves, previously maintained in culture for 11 days. This medium produces rapid neuron-like differentiation of cultured PC12 cells, as revealed by an increase in the size of the cell body and by the extension of short and/or long neurites by most of the cells. Neuregulin present in the conditioned medium was demonstrated to play a key role in the observed differentia… Show more

“…Studies show that neuronal differentiation is a very complex process, and several factors have been involved to induce neural precursors into functional differentiation. There is also strong evidence that neurotrophins and bFGF can induce neurogenesis in neural stem cell cultures [23,24]; therefore, we consider that the concomitant use of NGF and bFGF associated with conditioned media can promote neuronal fate in 3D culture. We found an increase in some transcription factors important for neuron specification, such as nestin, Map2, and β3-tub after 3 weeks of exposure to neural medium.…”

“…The anti‐oxidant and anti‐thrombotic effects of Acorus species have been compared (Choi et al ., ; Bang et al ., ). PC12 cell is a neuron‐like cell culture model (Villegas et al ., ), which could respond to nerve growth factor (NGF) in inducing the outgrowth of neurite, as well as the expression of neurofilament (Zhou et al ., ). It is a common model for testing the event of neuronal differentiation.…”

“…It is obvious that neuronal differentiation is a very complex process regulated by interplay among intrinsic cellular programs; cell‐cell and cell‐substrate interactions and several factors have been involved to induce neural precursors. There is also definitive evidence that neurotrophins and growth factors can induce neural differentiation in stem cell cultures . Neuroblastoma cells secrete many secretary factors may have the properties of other neurotrophic factors such as RA or NGF that can switch on the differentiation cascade of events.…”
