“…All key events within the niche—maintaining the pool of NSCs, their recruitment and proliferation, maturation of NPCs, migration and differentiation of neurobalsts—are driven by the changes in the tissue stiffness and local microenvironment which is provided by the secretory activity of resident cells or by the permeability of the BBB (either in SGZ or SVZ), BCSFC and VB (in SVZ). 28 , 35 , 235 , 236 …”

“…Hence, the expression of light-activated molecules in NSKs under the GFAP promoter can regulate their activity and, accordingly, regulate neurogenesis. For example, the optogenetic activation of ChR2 in astrocytes within an artificial neurogenic niche model has been demonstrated to stimulate neurogenesis [165]. However, it is important to note that there is conflicting evidence suggesting that astrocytes that do not express GFAP and vimentin (GFAP−/−Vim−/−) may have an inhibitory effect on neurogenesis and neuronal differentiation.…”

“…Less is known about the application of optogenetics for controlling astroglia-driven regulation of adult neurogenesis. We have demonstrated before that optogenetic stimulation of niche astrocytes expressing channelrhodopsin-2 under the GFAP promoter was efficient in activating the neurogenic potential of NSCs/NPCs in the in vitro neurogenic niche model or in implanted intrahippocampal neurospheres ex vivo in experimental Alzheimer's disease [184,185]. Photostimulation of iPSC-derived ChR2-expressing astrocytes co-cultured with iPSC-derived neurons results in effective bidirectional signaling, which is important for supporting the maturation of neurons and the establishment of a functional synaptic network, even though the transcriptomic analysis confirms that iPSCsoriginated astrocytes are relatively immature compared to adult cortical astrocytes [186].…”
