Neurochondrin Negatively Regulates CaMKII Phosphorylation, and Nervous System-specific Gene Disruption Results in Epileptic Seizure

Abstract: Neurochondrin is a novel cytoplasmic protein and possibly involved in neurite outgrowth, chondrocyte differentiation, and bone metabolism. Our previous trial in disclosing its role by the loss of function in mice failed because of the lethality in utero. In this study, we eliminated the neurochondrin gene expression preferentially in the nervous system by the conditional knockout strategy. Our results showed that neurochondrin is a negative regulator of Ca 2؉ /calmodulin-dependent protein kinase II phosphoryla… Show more

“…6B and 7B]). Other genes that remained downregulated during infection were neurochondrin (NCDN), involved in neurite outgrowth (40); complexin 1 (CPLX1), expressed by neurons and regulating synaptic vesicle exocytosis (41) (confirmed by IHC and qRT-PCR [ Fig. 6C and 7C]); and progestin and adipoQ receptor 6 (PAQR6) a G-coupled membrane progesterone receptor that exhibits neuroprotective properties (42) (Fig.…”

“…Given that Rab5 is required for internalization of AMPAR during long term depression (52), Ncdn might regulate internalization of AMPAR, directly acting on early endosome dynamics or indirectly modulating a signal through mGluR5. Because conditional Ncdn KO mice showed apparent pathological phenotypes (epileptic seizures and schizophreniarelevant behaviors) (23,24), genetic experiments using knock-in mice in which Ncdn palmitoylation-deficient mutant is expressed should reveal the roles of Ncdn palmitoylation in these pathophysiological phenotypes.…”

“…GPCR, G-protein coupled receptors predominantly expressed in the brain, has a very high CSSPalm score (Fig. 1C, Table 1, and supplemental Table S2), and plays an important role in neurite outgrowth (20,48) and synaptic plasticity (20,23,24). Thus, we subsequently focused on Ncdn palmitoylation.…”
