Neural differentiation of transplanted neural stem cells in a rat model of striatal lacunar infarction: light and electron microscopic observations

Abstract: The increased risk and prevalence of lacunar stroke and Parkinson's disease (PD) makes the search for better experimental models an important requirement for translational research. In this study we assess ischemic damage of the nigrostriatal pathway in a model of lacunar stroke evoked by damaging the perforating arteries in the territory of the substantia nigra (SN) of the rat after stereotaxic administration of endothelin-1 (ET-1), a potent vasoconstrictor peptide. We hypothesized that transplantation of neu… Show more

“…Experiments were performed at 2-3 months and 6 months-old female mice. Mice were processed according to standard procedures described previously (Muneton-Gomez et al, 2012). Briefly, after brain fixation with 4% (w/v) paraformaldehyde in 0.1M PB, tissue was cryopreserved in 30% sucrose in 0.1M PB and cut into blocks.…”

“…Grafted neurons can incorporate into the neuronal circuitry and form synaptic connections with host neurons. 4 , 5 , 6 , 7 , 8 However, the extent to which grafted neurons can integrate into existing circuits is not clear. A key question is whether grafted neurons will receive orthotopically afferent inputs from host neurons and function properly.…”

“…30 Briefly, at the end of the treatment, the brains were fixed by transcardial perfusion with 100 mL 4% (w/v) paraformaldehyde in 0.1 M phosphate buffer (PB), pH 7.4. Afterwards, brains were re moved, cut into blocks and fixed in the same fixative for 4 h at room temperature.…”
