Neural crest cells development and neuroblastoma progression: Role of Wnt signaling

Abstract: Neuroblastoma (NB) is one of the most common heterogeneous extracranial cancers in infancy that arises from neural crest (NC) cells of the sympathetic nervous system. The Wnt signaling pathway, both canonical and noncanonical pathway, is a highly conserved signaling pathway that regulates the development and differentiation of the NC cells during embryogenesis. Reports suggest that aberrant activation of Wnt ligands/receptors in Wnt signaling pathways promote progression and relapse of NB. Wnt signaling pathwa… Show more

“…According to our KEGG results, the DEGs identified were mainly enriched in the Wnt signaling pathway, which has been shown to play vital roles in epidermal development, stem cell self-renewal, and wound healing [ 55 , 56 , 57 , 58 , 59 , 60 , 61 ]. The horns of ruminants have a common cellular origin from neural crest cells [ 3 ] and the Wnt signaling pathway is essential for regulating the fate, migration, and proliferation of cranial neural crest cells [ 62 , 63 , 64 , 65 ]. In addition, antler regeneration may be influenced by the Wnt signaling pathway [ 37 ].…”

“…This cleavage process produces CTF2, facilitated by γ-secretase, enhancing the transcriptional activity of β-catenin and cyclin D1. Consequently, this inhibits the G1/S transition, ultimately inducing the delamination of NCCs [31]. However, the connection between BMPs and WNT is more complex.…”

“…There are studies that describe how the highly conserved Wnt signaling pathway regulates the development and differentiation of the neural crest cells during embryogenesis. When Wnt signaling pathway is inhibited, it induces apoptosis and hinders proliferation and oncogenesis in all types of neuroblastic cells [ 43 ]. BORIS is a DNA-binding protein that recruits certain transcription factors and induces tumorigenesis.…”
