Network Pharmacology to Explore the Molecular Mechanisms of Prunella vulgaris for Treating Hashimoto’s Thyroiditis

Abstract: Purpose:Prunella vulgaris (PV), a traditional Chinese medicine, has been used to treat patients with thyroid disease for centuries in China. The purpose of the present study was to investigate its bioactive ingredients and mechanisms against Hashimoto’s thyroiditis (HT) using network pharmacology and molecular docking technology to provide some basis for experimental research. Methods: Ingredients of the PV formula were retrieved from the Traditional Chinese Medicine Systems Pharmacology (TCMSP) database. “…Edaravone is a drug that scavenges hydroxyl radicals [104], it acts on the STAT3/PI3K/Akt pathway to effectively improve autoimmune thyroiditis and has become an emerging drug for the treatment of HT [104]. PVmediated HMGB1 inhibition decreased the expression of pro-inflammatory cytokines and suppressed the HMGB1-TLR9 signaling pathway in, while downregulating the proportion of Th1, Th2 and Th17 cells in splenocyte, provides a potential therapeutic value for HT [81], the lignan component of PV has a strong affinity for the disease protein of HT, and quercetin has a strong affinity for serum thyroid peroxidase (TPO), further confirming that PV can effectively treat HT [100].…”

“…The most common way to control HT is to take levothyroxine (L-T4) to control the disease [100]. Longterm use of 1.6-1.8 mg per kg to achieve normal levels of thyrotropin in the body [21].…”

“…What’s more, there are few reports on the PI3K-Akt signaling pathway in HT. The literature suggests that Prunella vulgaris may play an anti-inflammatory role by regulating the PI3K-Akt signaling pathway, thus realizing the treatment of HT ( 39 ). but this study only draws the conclusion through bioinformatics analysis, and has no in vivo or in vitro evidence.…”

“…R-language packages ‘ClusterProfiler' and ‘ http://org.Hs.eg.Db ' were used to analyze the enrichment of GO and KEGG to explore the underlying biological function (BP), cellular components (CC), and molecular function (MF) and signaling pathways. A p .adjust (FDR) < 0.05 was considered statistically significant [ 21 ]. The bubble chart and Circro circles for GO and KEGG results were visualized by the Sangerbox platform ( http://sangerbox.com/Tool ).…”
