Network pharmacology-based strategy to investigate pharmacological mechanisms of Zuojinwan for treatment of gastritis

Abstract: BackgroundZuojinwan (ZJW), a classic herbal formula, has been extensively used to treat gastric symptoms in clinical practice in China for centuries. However, the pharmacological mechanisms of ZJW still remain vague to date.MethodsIn the present work, a network pharmacology-based strategy was proposed to elucidate its underlying multi-component, multi-target, and multi-pathway mode of action against gastritis. First we collected putative targets of ZJW based on TCMSP and STITCH databases, and a network contain… Show more

“…With the rapid development of bioinformatics, network pharmacology has emerged as a powerful tool to explore TCM (Cao et al, 2018;Yu et al, 2018). Based on system-biology, multi-directional pharmacology, and highthroughput analysis, network pharmacology can thoroughly explain the complicated relationship between drugs and diseases by constructing biological network and network visualization analysis of the potential active ingredients, hub targets, signaling pathways and diseases (Yuan et al, 2017;Ning et al, 2018;Huang et al, 2019).…”

“…Because most compounds in Chinese medicine have poor pharmacological properties, they cannot effectively bind to specific cell protein targets. Thus, many researchers have recommended that the OB ≥ 30% and DL ≥ 0.18 molecules be considered to have better pharmacological effects and be selected as candidate compounds for further analysis (Chen G. et al, 2014;Song et al, 2018;Yu et al, 2018;Kim et al, 2019;Ma et al, 2019;Yang et al, 2019). This study also used this principle to screen compounds for further analyses.…”

“…By using web-based approaches, systems pharmacology can systematically determine the effects and mechanisms of drugs used to treat complex diseases at the molecular, cellular, tissue, and biological levels. This research strategy for Danlu capsules, the Maxing Ganshi Decoction, the Wei Pi Xiao Decoction, Zuojinwan, and other capsules from traditional Chinese medicine has been widely adopted (Huang et al, 2017;Song et al, 2018;Yu et al, 2018;Yang et al, 2019).…”

“…Modern pharmacological research (Shin and Seo, 2017) indicates that TCM decoctions can effectively lower the expression of inflammatory proteins in rats with reflux disease and also lower inflammatory mediators and the activation of cytokines nuclear factor-kappa (NF-kB) by protein expression related to the mitogenactivated protein kinase (MAPK) signaling pathway. In a network pharmacology study of ZJP (Yu et al, 2018), it was found to be mainly involved in anti-inflammatory effects. For the treatment of gastric symptoms, the conclusions indicate that it mainly passed through potential targets such as EGFR, IL-6, IL-1b, TNF-a, and MCP-1, and known CCKBR and IL-12b targets and a link target, NF-kB, to inhibit inflammatory reactions.…”
