Network Analysis and Human Single Cell Brain Transcriptomics Reveal Novel Aspects of Alpha-Synuclein (SNCA) Biology

Abstract: Alpha-synuclein (SNCA) aggregates are pathological hallmarks of synucleinopathies, neurodegenerative disorders including Parkinson's Disease (PD) and Lewy Body Dementia (LBD). Functional networks are not yet well-characterized for SNCA by CNS cell type. We investigated cellspecific differences in SNCA expression using Allen Brain Database single-nucleus RNA-seq data from human Middle Temporal Gyrus (MTG, 15,928 nuclei) and Anterior Cingulate Cortex (ACC, 7,258 nuclei). Weighted gene co-expression analysis (WGC… Show more

“…We identified five types of neurons; PPP1R1B + /DRD1 + medium spiny projection neurons (D1MSN); PPP1R1B + /DRD2 + medium spiny projection neurons (D2MSN); a distinct spiny neuronal population (MSN) characterized by expression of PPP1R1B + /ADARB2 + ; and two GAD1 high /GAD2 high GABAergic inhibitory neuronal clusters (GABA1 and GABA2), as shown in Figure 2A. We did not find any SLC17A7 + glutamatergic/excitatory neurons, which are abundant in cortical areas [11].…”

“…Oligodendrocyte lineage nuclei were then subset from the integrated objects, to undergo reclustering and profiling to identify cell subtypes, evaluate their relationships by pseudotime trajectory analysis, and identify differentially expressed genes between disease (PD or MSA) and Control nuclei. Among oligodendrocyte lineage nuclei, oligodendrocyte precursor cells (OPC; markers: PDGFRA/VCAN), committed oligodendrocyte precursor cells (COP), and multiple clusters of oligodendrocytes (MOG/MAG/OPALIN/HAPLN2) have been described in previous studies [4,15]. Re-clustering of the OPC/Oligodendrocyte lineage cells was performed based on the rationale that unsupervised clustering of broad cell types might generate different cluster separations among oligodendrocyte subtypes than might be identified through applying the procedure only to oligodendrocyte lineage cells.…”

“…In contrast, MSA occurs at a much lower frequency than PD, with an estimated mean incidence rate of 0.6-3 per 100,000 people [2,3]. In both PD and MSA, intracytoplasmic inclusions of α-synuclein are observed on post-mortem histologic examination, although the predominant cellular localization of α-synuclein differs between the two conditions: α-synuclein aggregates are observed mainly as neuronal intracellular aggregates (Lewy bodies) in PD [4,5] while in MSA, these aggregates occur most frequently as oligodendroglial cytoplasmic inclusions [5,6].…”
