Network analysis, and human and animal studies disclose the anticystitis glandularis effects of vitamin C

Abstract: Background Our present study aimed to unravel the therapeutic biotargets of vitamin C (VC) against cystitis glandularis (CG), and to elucidate the molecular mechanisms for VC treating CG. Methods Network pharmacology was used to predict therapeutic targets of VC against CG, and to identify molecular mechanisms. In addition, further human and animal studies were designed to validate the bioinformatic findings through biochemical tests, computerized tomography scans, and immunostaining assays. “…The R packages of ‘ClusterProfiler’, ‘ReactomePA’ and ‘AnnotationHub’ were used for enrichment analysis of Gene Ontology (GO) in biological function (BP) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway using the previously identified best targets. This data were used to generate a corresponding bubble chart and histogram [ 26 ]. GO and KEGG analyses were performed using clusterProfiler (version 3.14.3) [ 27 ] and ReactomePA (version 1.30.0) [ 28 ] from the R packages.…”

“…The Animal Guidelines for the Care and Use of Animals at Guilin Medical University were approved. After acclimatization for approximate 7‐day, we prepared the CG mice chemically using our previous method 13,14 . And then CG mice were treated intragastrically with 20, 40 mg/kg PA each day for lasting 14‐day.…”

“…Encouragingly, our previous reports show that some potential anti‐CG agents, including glycyrrhetinic acid, vitamin C, are found with potential pharmacological actions against CG. The underlying mechanisms of glycyrrhetinic acid against CG are associated with inhibition of cell death and inflammatory stress, while the anti‐CG mechanism of vitamin C is related to reduction of intracellular tumor necrosis factor (TNF)‐α, IL6, and c‐Jun expressions 13,14 . Currently, the association between PA and CG is uninvestigated.…”
