Nesfatin-1 Regulates Feeding, Glucosensing and Lipid Metabolism in Rainbow Trout

Abstract: Nesfatin-1 is an 82 amino acid peptide that has been involved in a wide variety of physiological functions in both mammals and fish. This study aimed to elucidate the role of nesfatin-1 on rainbow trout food intake, and its putative effects on glucose and fatty acid sensing systems. Intracerebroventricular administration of 25 ng/g nesfatin-1 resulted in a significant inhibition of appetite, likely mediated by the activation of central POMC and CART. Nesfatin-1 stimulated the glucosensing machinery (changes in… Show more

“…20 This, along with the fact that nesfatin-1 can cross the blood-brain barrier, 21,22 suggests that nesfatin-1 may have pleiotropic effects. In addition to maintaining the feeding balance, this peptide plays an important role in glucose homeostasis, 23 lipid metabolism, 24 modulation of gastrointestinal functions, 25 cardiovascular, 9 and reproductive functions. 26 It may also be involved in epilepsy, 27 psychological disorders, including stress, 28 sleep disorders, 29 anxiety, 30 and depression.…”

“…Nesfatin-1 stimulates fatty-acid oxidation by activating AMP-activated protein kinase in diabetic rats [33], chronic subcutaneous infusion of nesfatin-1 reduced plasma cholesterol and triglyceride and elevate HDL-c levels in other animal models [21,34]. Central nesfatin-1 in the brain may reduce the lipogenic activity and enhance fatty acid oxidation in the rainbow trout liver [35]. Theoretically, anti-atherogenic effect of nesfatin-1 may be mediated via by its anti-oxidant and anti-inflammatory properties and its enhancing effect on insulin sensitivity [4,5,36].…”

“…Over the last decade, significant information on the biological actions of nesfatin-1 has been reported in vertebrates, including fish. The main physiological roles of nesfatin-1 in fish include regulation of food intake [1,2], pituitary and gonadal hormone synthesis and/or secretion [3][4][5], cardiovascular functions [6], and oocyte maturation [5]. Research using zebrafish in our lab identified a nesfatin-1-like peptide (Nlp), which is processed from a precursor nucleobindin-1 (NUCB1) that has very high amino acid sequence similarity with NUCB2 [7].…”
