Nerve growth factor increases MMP9 activity in annulus fibrosus cells by upregulating lipocalin 2 expression

Abstract: Purpose Nerve growth factor (NGF) expression and activity is important in chronic lower back pain but may also act as a pro-catabolic factor in the pathogenesis of intervertebral disc (IVD) degeneration. Lipocalin 2 (Lcn2) expression in IVD was upregulated by NGF stimulation in our previous study. The current study was undertaken to identify potential mechanisms of the latter effect including potential interactions between Lcn2 and matrix metalloproteinase 9 (MMP9). Methods Rat annulus fibrosus (AF) cells were… Show more

“…Kao and colleagues demonstrated that nerve growth factor (NGF), an IVDD inducer, up-regulates the expression of LCN2, which forms covalent complexes with MMP9, blocking its auto-degradation and hence, increasing its activity, in rat AF cells [69,70]. In fact, NGF increased the expression of MMP9 protein but not gene expression in AF cells [69]. These results indicated that LCN2 could be implicated in disc degeneration, but further studies are needed.…”

“…This could be explained by an inappropriate model for the detection of MMP2/9, the intracellular gelatinases MMPs are normally secreted in the extracellular compartment for a variety of cell lineages [40][41][42]. Therefore, it is more relevant to assess the incorporated gelatinases in culture supernatant rather than within the cells lysates [43] as also detected previously for IVD cells [44].…”

“…A possible reason may be that extracellular free-NGF is degraded rapidly by metalloproteinase (MMP)-9 [34]. MMP-9 is abundant in VLU exudate [35, 36], and NGF itself promotes expression and activation of MMP-9 [37, 38]. NGF also activates neutrophils, which may contribute to the prolongation of inflammation in VLU and are the primary MMP-producing cell [2].…”
