Nephroprotective effect of umbelliferone against cisplatin‐induced kidney damage is mediated by regulation of NRF2, cytoglobin, SIRT1/FOXO‐3, and NF‐kB‐p65 signaling pathways

Abstract: Cisplatin (Cis) is one of the most potent and effective broad‐spectrum antitumor drugs, but its use is limited due to nephrotoxicity. The current study investigated the renoprotective effect of umbelliferone (UMB) on Cis‐induced nephrotoxicity in rats. Renal injury was induced by a single injection of Cis (7 mg/kg, ip). Our results exhibited that the injection of Cis significantly disrupted renal function biomarkers as well as KIM‐1 expression. The expressions of TNF‐α, IL‐1β, NF‐kB‐p65, and IKKβ were elevated… Show more

“…These results confirm the regulatory role of SIRT-1 in drug-induced renal injuries [25]. While the modulatory effect of cisplatin of SIRT-1 was previously reported [9][10][11]26,27], this work is the first, to the best of our knowledge, to report the effect of quinacrine on SIRT-1 regulation.…”

“…Furthermore, umbelliferone is a coumarin derivative that was extracted from C. sativum using methanol [ 111 ] and is also found to increase the expression of Nrf2 in addition to CREB, SIRT1, FOXO-3, PPAR-γ genes, and proteins. SIRT1 upregulation plays a key role in regulating oxidative defense mechanisms and DNA repair [ 112 ] and can be considered one of the pathways in which coriander illustrates its antioxidant and anti-atherogenic effects.…”

“…The well-reported pathogenesis of CIS toxicity is related to reactive oxygen species (ROS)mediated pathways, including oxidative injury and inflammatory perturbations [39,40]. Redox impairment provokes oxidative stress and aberrant signalling for tissue injury.…”
