Neovascularization Potential of Blood Outgrowth Endothelial Cells From Patients With Stable Ischemic Heart Failure Is Preserved

Abstract: BackgroundBlood outgrowth endothelial cells (BOECs) mediate therapeutic neovascularization in experimental models, but outgrowth characteristics and functionality of BOECs from patients with ischemic cardiomyopathy (ICMP) are unknown. We compared outgrowth efficiency and in vitro and in vivo functionality of BOECs derived from ICMP with BOECs from age‐matched (ACON) and healthy young (CON) controls. Methods and ResultsWe isolated 3.6±0.6 BOEC colonies/100×106 mononuclear cells (MNCs) from 60‐mL blood samples of… Show more

“…4 There is growing evidence that endothelial progenitor cells (EPCs) are involved in myocardial angiogenesis of ICM patients. 5 However, in contrast to EPCs from healthy subjects, the transplantation of EPCs from CHF patients failed to increase angiogenesis. This reveals that impaired EPC angiogenesis occurs in CHF patients.…”

“…It has been reported that LEPCs play a role in wound healing, with the greatest angiogenic potential . Clinical studies have also shown that LEPCs improve vascular outcomes in ischemic and chronic wound patients .…”

“…After homing, activated LEPCs initiate adhesion to vascular endothelial cells (ECs). In sequence, transendothelial migration into the sites of the wounded endothelium and differentiation into mature ECs proceed . Throughout this process, LEPCs promote endothelial remodeling and wound healing by supplying neovasculature.…”

“…There is evidence regarding potent possibilities to improve the EPCs' capacity using various methods, i.e. aerobic exercise, lipid lowering drugs, ACE inhibitors, calcium channel blockers, antidiabetic drugs including metformin, sitagliptin [71][72][73][74][75]. Several mechanisms are involved in the attenuation of EPCs functionality by mentioned above approaches, i.e.…”
