Post-traumatic stress disorder (PTSD) is twice as common in women as in men; it is a major public health problem whose neurobiological basis is unknown. In preclinical studies using fear conditioning and extinction paradigms, women and female animals with low estrogen levels exhibit impaired extinction retrieval, but the mechanisms that underlie these hormone-based discrepancies have not been identified. There is much evidence that estrogen can modulate dopaminergic transmission, and here we tested the hypothesis that dopamine-estrogen interactions drive extinction processes in females. Intact male and female rats were trained on cued fear conditioning, and received an intraperitoneal injection of a D1 agonist or vehicle before extinction learning. As reported previously, females that underwent extinction during low estrogen estrous phases (estrus/metaestrus/diestrus (EMD)) froze more during extinction retrieval than those that had been in the high-estrogen phase (proestrus; PRO). However, D1 stimulation reversed this relationship, impairing extinction retrieval in PRO and enhancing it in EMD. We also combined retrograde tracing and fluorescent immunohistochemistry to measure c-fos expression in infralimbic (IL) projections to the basolateral area of the amygdala (BLA), a neural pathway known to be critical to extinction retrieval. Again we observed diverging, estrous-dependent effects; SKF treatment induced a positive correlation between freezing and IL-BLA circuit activation in EMD animals, and a negative correlation in PRO animals. These results show for the first time that hormone-dependent extinction deficits can be overcome with non-hormone-based interventions, and suggest a circuit-specific mechanism by which these behavioral effects occur.

Stress in Neonatal Rats with Different Maternal Care Backgrounds: Monoaminergic and Hormonal Responses

The first 2 weeks of life in rats are known as the stress hyporesponsive period because stress responses in pups are diminished as compared to adult animals. However, it is considered a critical period in development in which infant rats are susceptible to environmental events, such as stressful stimuli and quality of maternal care received. These early life events have long-lasting effects, shaping a variety of outcomes, such as stress responsivity. This study investigated the effects of maternal care and sex differences on the response to an aversive stimulus in rat pups from high (HL) and low licking (LL) mothers. Plasma corticosterone, oxytocin (OT), and central monoaminergic activity in 13-day-old rats submitted to cold stress were analyzed. Stress increased plasma corticosterone and marginally decreased hypothalamic dihydroxyphenylacetic acid/dopamine ratio. HL pups showed higher levels of plasma OT than LL pups. The maternal effect was also detected in the hippocampus, in which 5-hydroxyindole-3-acetic acid/serotonin ratio was increased in HL pups, independently of the sex and stress. Investigating the early life events is useful not only into understand the neurobiological and hormonal mechanisms underlying maternal and stressful influences on infant development into a healthy or psychopathological adult phenotype, but also to unveil the immediate outcomes on infancy.

The objective of this study was to assess the role of environmental/nest components and maternal behavior after several neonatal interventions on subsequent behavioral responses. Male Wistar rats were subjected to different neonatal interventions and were later evaluated for innate fear-like behavior in adulthood. The experimental groups included nonhandled (i.e., animals were not touched), handled (i.e., animals were separated from their mother, removed from the nest, and handled for 10 min/day), brief maternal separation (i.e., the mother was removed from the homecage for 10 min/day, and the pups remained in their cages without being touched), and tactile stimulation (i.e., the mother was removed from the homecage, and pups were stimulated with a brush for 10 min/day within the nest). The mother's behavior was recorded during the neonatal period, and the male pups were later tested in the open field as adults. The results revealed that only mothers whose pups were handled had an increase in the duration of licking behavior compared with the other groups. In the open field test, we observed decreased behavioral innate fear-like responses in male adults in the handled and brief separation groups compared with the others groups. Our results confirm that interventions during the neonatal period cause stable behavioral changes (decreased innate fear) in adulthood and that absent or excessive tactile stimulation appears to be an important factor. Both repeated disruption of the mother-infant relationship and withdrawal from the environment/nest are factors that exert profound effects on the development of the animals.
