Negative ion ‘chip‐based’ nanospray tandem mass spectrometry for the analysis of flavonoids in glandular trichomes of Lychnophora ericoides Mart. (Asteraceae)

Abstract: This paper reports a method for the analysis of secondary metabolites stored in glandular trichomes, employing negative ion 'chip-based' nanospray tandem mass spectrometry. The analyses of glandular trichomes from Lychnophora ericoides, a plant endemic to the Brazilian 'cerrado' and used in traditional medicine as an anti-inflammatory and analgesic agent, led to the identification of five flavonoids (chrysin, pinocembrin, pinostrobin, pinobanksin and 3-O-acetylpinobanksin) by direct infusion of the extracts of… Show more

“…The flavonoid aglycones as pinobanksin, chrysin, pinocembrin and pinostrobin displayed different tissue distribution compared to vicenin-2, being identified in the abaxial stomatic crypts of L. ericoides (Fig. 8) inside the head of glandular trichomes, which confirms previous observations17.…”

“…The flavonoid aglycones (pinobanksin, chrysin, pinocembrin and pinostrobin) have been identified in the abaxial stomatic crypts of L. ericoides inside the head of glandular trichomes, confirming previous observations17. A number of flavonoid aglycones with UV protection functions have been detected in the cuticle19; however, these compounds were not identified in the cuticles of the species studied here.…”

“…Chromatographic peaks 2 and 3 ( Figure 2) were identified as the di-C-glucosylflavones 6,8-di-C-b-glucosylapigenin (vicenin-2) and 6,8-di-C-b-glucosylchrysin, respectively, due to the co-elution with standards of these compounds previously isolated from L. ericoides, as stated in GobboNeto et al 31 Their measured accurate masses are in accordance with the calculated exact molecular masses of the respective protonated molecules, and the fragmentation Differential Metabolic and Biological Profiles of Lychnophora ericoides Mart. “…STL in Asteraceae are mainly sequestered in glandular trichomes. 31,37 To exclude the possibility that the observed difference in STL presence is related to a differential capacity of developing glandular trichomes, leaves of all the analysed plants were examined under a dissecting microscope. Capitate glandular trichomes were present in similar amounts and sizes in leaves of all analysed plants.…”

“…The MS/MS spectra were compared with literature reports for chlorogenic acids [23], [24], flavonoids [25], [26], and sesquiterpene lactones [24]. The annotation of chromatographic peaks was performed partially based on previous phytochemical studies of Vernonia species.…”
