Miniaturization is a key technological approach to realize high performance separation systems that have several important features required in modern analytical methods. A smaller scale of the stationary phase synthesis enables a systematic analysis of the retention behavior and also a more efficient development of novel stationary phases in liquid chromatography. Development of microscale sample preparation technique has been regarded as a valuable supporting approach to the development of microscale separation methods. Effective on-line coupling of the microscale sample preparation and separation allows an efficient and rapid analysis without a loss of sensitivity for most of the analytical separations. In this review, development of the miniaturized separation techniques and the applications have been described from the view point of the authors' research group, including the development of novel stationary phase, microscale sample preparation techniques for liquid and gas sample matrices.

Spherical Polyimide Particles as a Novel Stationary Phase in Liquid Chromatography

Spherical polyimide (PI) particles have been introduced as a stationary phase in liquid chromatography (LC). The PI was prepared with the starting materials of pyromellitic dianhydride (PMDA) and 4,4'-diaminodiphenyl ether (4,4'-oxydianiline, ODA), resulting the spherical PI (PMDA-ODA type) particles with an average diameter of about 5 μm. The synthesized PI particles were packed into a conventional blank LC column having a typical size of 4.6 mm i.d, 150 mm length, and the retention behavior for a group of polycyclic aromatic compounds (PACs) was evaluated in LC. The retention behavior was compared with that obtained on a commercially-available octadecylsilica (ODS) phase. The results indicated a significantly larger retentivity of PACs on the PI stationary phase than that on typical ODS stationary phases, where the retention tendency on the PI phase for various PACs having different molecular size and shape was similar to the trend on the ODS phase. The large retention on the PI phase can be interpreted on the basis of the surface structure of the PI particles, because a number of aromatic functionalities are in the chemical structure of the polymer chain backbone.

Analysis of Biogenic Volatile Organic Compounds in Air Using a Solid-Phase Extraction-Type Collection Device

In this study, the quantitative determination of gaseous biogenic volatile organic compounds (BVOCs) including monoterpenes and sesquiterpenes was accomplished using a solid-phase extraction-type collection device. The collection device was fabricated by packing styrene-divinylbenzene polymer particles into a specially designed glass cartridge. The retention performance of the collection device for BVOCs was quantitatively evaluated at 35°C with different volumes of air samples. The device showed good retention performance for the investigated BVOCs, that is, no breakthrough occurred for monoterpenes up to an air sampling volume of 150 L or for sesquiterpenes up to a sampling volume of 30,000 L. The elution performance was evaluated by passing organic solvents into the collection device, and an excellent elution recovery was obtained with 10 mL dichloromethane. Finally, the analytical method using the collection device was applied to determine the gaseous monoterpenes and sesquiterpenes from a grated carrot and the air inside a wooden house, and the results demonstrated the applicability of the method for the quantitative determination of BVOCs in several sample matrices.
