Nebulized perflubron and carbon dioxide rapidly dilate constricted airways in an ovine model of allergic asthma

Abstract: BackgroundThe low toxicity of perfluorocarbons (PFCs), their high affinity for respiratory gases and their compatibility with lung surfactant have made them useful candidates for treating respiratory diseases such as adult respiratory distress syndrome. We report results for treating acute allergic and non-allergic bronchoconstriction in sheep using S-1226 (a gas mixture containing carbon dioxide and small volumes of nebulized perflubron). The carbon dioxide, which is highly soluble in perflubron, was used to … Show more

“…Large-animal models can, however, play an important role in filling this need. Indeed, the physiological and immunological similarities between sheep and human lungs ( 60 , 61 ) have contributed to the widespread use of sheep as translational models for human lung research ( 26 , 27 ), including for asthma ( 62 65 ), the delivery of drugs via the upper respiratory tract ( 66 68 ), emphysema ( 69 71 ), pulmonary hypertension ( 72 74 ), physical lung injury ( 75 78 ), lung infection ( 28 , 79 81 ), respiratory distress syndromes ( 82 85 ), asbestosis ( 86 88 ), and lung cancer ( 89 , 90 ).…”

“…For this study, a sample size of 16 subjects was estimated to be able to detect a treatment difference of at least a 14% change in FEV 1 with a 95% confidence interval with a power of over 90% in a two-sided test, assuming that the variability observed would be like the response to S1226 in the sheep allergen challenge model [ 15 ]. The sheep model used airway resistance as a model of airway caliber.…”

“…They involve pH and epithelial-dependent mechanisms [ 13 ] and do not require adrenergic [ 14 ] or cholinergic pathways [ 12 ]. The combination of CO 2 with perflubron in preclinical studies produced a synergistic effect, providing rapid and sustained bronchodilation [ 13 15 ]. S1226 showed rapid opening of airways in sheep sensitized and challenged with house dust mites and rats sensitized and challenged with ovalbumin.…”

“…Sheep have been widely used in biomedical research to model a number of human pathologies including asthma (68,69), osteoporosis (70), sepsis (71) and acute lung injury (ALI) (72)(73)(74), as they also share many physiological similarities with humans (75)(76)(77).…”
