Near infrared light responsive hybrid nanoparticles for synergistic therapy

Abstract: A near infrared (NIR) light responsive chromophore 7-(diethylamino)-4-(hydroxymethyl)-2H-chromen-2-one (DEACM) was synthesized and incorporated to β-cyclodextrins with cRGD functionalized poly(ethylene glycol), the amphiphiles were coordinated with Au nanorods or nanoparticles to load anticancer drug doxorubicin (DOX) for fabricating hybrid nanoparticles. The π-π stacking interaction between DEACM and DOX was formed in the hybrid nanoparticles, which contributed to the high drug loading content. The Au nanorod… Show more

“…Then, the dialysis tube was immersed into a 50 mL tube containing 20 mL dextrose solution (fraction 5%) in a shaker at 37°C and 120 rpm. At predetermined times (5,10,15,20,30,45, and 60 min), 5 mL of the external dextrose solution was replaced with the corresponding fresh dextrose solution. The concentration of Gd(III) was determined by ICP-AES, and I -was measured using the chloric acid method.…”

“…13 Upon near-infrared (NIR) light laser irradiation, the dissipation of absorbed light energy as heat can not only mediate hyperthermia of tumor with high spatial and temporal resolution but also activate the rapid release of contents at the site of interest. [14][15][16][17] The introducing of the light-triggered release systems in CA delivery will help to utilize the self-quench property of fluorescence at high concentrations, and has been used for optic imaging. 18 However, previous reports on light-triggered nanotheranostics mostly focused on cancer treatment.…”

“…These results further proved that DOX-loaded gels had better inhibition effect on tumours. Metastasis and invasion always go with angiogenesis in the process of growth for solid tumours [46]. In order to research the tumour suppression mechanism and the antitumour efficacy, the Ki-67 and CD31 antigen staining were evaluated to assess the antitumour efficacy on tumour cell proliferation, and TUNEL assay was performed to research the apoptotic programmed cell death as shown in Figure 8.…”

“…Cytotoxicity assay. The biocompatibility of probes in living cells is an important consideration (30,31). In the current study, the ICG and IR787 probes were evaluated in vitro to determine any cytotoxicity in A549 cells using the MTT method.…”
