The two functional groups of -OH and -NH2 of chitosan monomer interacted with functional groups of -Ac dan -NH2 of amino acid of A, C, T, dan P in ADTC1 peptide which results in the binding energy. The interaction between Ac-CA-NH2 and Ac-TP-NH2 peptide and chitosan dimer was studied. The purpose of this study to determine the energy of interaction between Ac-CA-NH2 and Ac-TP-NH2 peptide with chitosan dimer segment. The energy of interactions were calculated by ab initio quantum method at the level of theory and basis set HF/ 6-31G(d,p). The interaction energy of configuration-1 and configuration-2 between chitosan dimer segment with Ac-CA-NH2 peptide were -26.144 kJ mol -1 and -66.5296 kJ mol -1 , respectively, with the average length of H-bonding was 2.010 Ǻ. Meanwhile, the interaction energy of configuration-1 and configuration-2 between chitosan dimer segment with Ac-TP-NH2 peptide were -41.5616 kJ mol -1 and -54.7272 kJ mol -1, respectively, and the average length of H-bonding was 2.053 Ǻ The conclusion was that the chitosan dimer segment has a moderate and different affinity of H-bonding with Ac-CA-NH2 and Ac-TP-NH2 peptide which was predicted as a basic principle of system delivery of ADTC1 and Chitosan.A b s t r a k Kata Kunci: drug delivery, peptida , kuantum ab inito, ADTC1 Salah satu faktor yang mempengaruhi sistem drug delivery dengan kitosan adalah kekuatan binding antara kitosan dengan ADTC1. Interaksi antara molekul kitosan dengan ADTC1 adalah melalui gugus -OH dan -NH2 pada monomer kitosan dengan gugus -Ac dan -NH2 pada monomer asam amino A, C, T, dan P pada peptida ADTC1. Telah dipelajari interaksi antara molekul Ac-CA-NH2 dan Ac-TP-NH2 penyusun peptida ADTC1 (Ac-CADTPPVC-NH2) dengan dimer kitosan. Konfigurasi interaksi yang lebih disukai atau lebih stabil dapat diperkirakan melalui interaksi komponen penyusun kitosan dan komponen penyusun peptida ADTC1 tersebut. Penelitian dibatasi pada dimer kitosan dan peptida Ac-CA-NH2 dan Ac-TP-NH2. Tujuan penelitian adalah untuk menentukan energi interaksi antara peptida Ac-CA-NH2 dan Ac-TP-NH2 dengan segmen dimer kitosan. Penelitian dilakukan dengan metode kuantum ab initio pada tingkat teori dan basis set HF/6-31 G(d,p). Hasilnya adalah energi interaksi konfigurasi-1 dan konfigurasi-2 antara dimer kitosan dengan peptida Ac-CA-NH2 masingmasing adalah -26,144 kJ mol -1 dan -66,5296 kJ mol -1 dengan panjang ikatan-H yang terbentuk rata-rata 2,010 Ǻ. Abstract:The aim of this work was to investigate the possibility of covalent cross-linker free, polyelectrolyte complex formation at the nanoscale between alginic acid (as sodium alginate, ALG) and protamine (PROT). Optimisation of the self-assembly conditions was performed by varying the type of polymer used, pH of component solutions, mass mixing ratio of the components as well as the speed and order of component addition on the properties of complexes. Homogenous particles with nanometric sizes resulted when an aqueous dispersion of ALG was rapidly mixed with a solution of PROT. The polyelectrolyte complex between ALG and PROT was confirmed by infrared spectroscopy. To facilitate incorporation of drugs soluble at low pH, pH of ALG dispersion was decreased to 2, however no nanoparticles (NPs)were formed upon complexation with PROT. Adjusting pH of PROT solution to 3 resulted in the formation of cationic or anionic NPs with a size range 70-300 nm. Colloidal stability of selected AAL/PROT formulations was determined upon storage at room temperature and in liquid media at various pH. Physical stability of NPs correlated with the initial surface charge of particles and was time-and pH-dependent. Generally, better stability was observed for anionic NPs stored as native dispersions and in liquids covering a range of pH.

Industrial Applications for Intelligent Polymers and Coatings

Bioactive polymers, by their structural confi guration and conformation, possess the ability to exert biological activities and consequently elicit responses from cells and tissues. Intelligent polymers are smart polymers which respond to internal and external stimuli in order to propel the release or modify the release of drugs. Natural polymers are biogenic, biocompatible, biodegradable, and safe for consumption. Consequently, they present as suitable materials that the human body can identify with and not treat as foreign bodies, thereby reducing the complications encountered when dealing with synthetic polymers. Natural polymers have been shown to be bioactive, exhibiting biological activities such as antitumor, anticoagulant, antioxidant, antimicrobial, antiulcer, anti-infl ammatory, and antirheumatic. In addition, natural polymers are meritorious materials for the fabrication of selfregulated or externally regulated drug delivery systems. These systems respond to the state of the environment for effi cacious therapy. Drug delivery technology is shifting from the controlled release of drugs over time to the release of drugs when and where needed, especially for chronic diseases. Indeed, intelligent polymers are choice polymers for such delivery systems. Their synthetic counterparts were actually synthesized to mimic these natural polymers which further buttress the need to revert to nature for intelligent and bioactive polymers. The contexts of natural bioactive and intelligent polymers have unique applications in drug delivery, embracing nanobiotechnology. This would ultimately benefi t drug delivery systems in benchmarking new drug formulations.
