Naturally anergic and suppressive CD25+CD4+ T cells as a functionally and phenotypically distinct immunoregulatory T cell subpopulation

Abstract: A CD4(+) T cell subpopulation defined by the expression levels of a particular cell surface molecule (e.g. CD5, CD45RB, CD25, CD62L or CD38) bears an autoimmune-preventive activity in various animal models. Here we show that the expression of CD25 is highly specific, when compared with other molecules, in delineating the autoimmune-preventive immunoregulatory CD4(+) T cell population. Furthermore, although CD25 is an activation marker for T cells, the following findings indicate that immunoregulatory CD25(+)CD… Show more

“…Despite the functional significance of CD25 and IL-2 signalling in naturally occurring Treg [11][12][13][14][15][16], the data presented here document deletion of diabetogenic cells by the fusion protein. Targeting cells that express the IL-2 receptor decreased the responsiveness to alloantigens in vitro, impaired adoptive transfer of the disease into NOD SCID mice and antagonised cyclophosphamide-induced destructive insulitis.…”

“…Third, FOXP3 + T cells in the pancreas and draining lymphatics are less sensitive to apoptosis induced by IL2-cas, resulting in expansion of C25 − FOXP3 + cells. Increased fractional expression of FOXP3 in the thymus, the origin of naturally occurring Treg [11,12] might be beneficial to restoration and maintenance of self-tolerance through resetting of the regulatory/effector equilibrium.…”

“…T reg cells must be stimulated to exert their suppressive effects; however, once stimulated, T reg cells act in an Ag-nonspecific manner (8,9,18,19). In vitro, the suppressive abilities of the T reg cells are connected to the anergic state, because addition of IL-2 not only breaks the anergy of the T reg cells but also abrogates their ability to suppress proliferation (20).…”

“…These assays were performed as described previously (20,35). Briefly, to assess proliferation, 2.5 ϫ 10 4 CD4 ϩ CD25 Ϫ or CD4 ϩ CD25 ϩ T cells (electronically sorted to a purity of Ͼ95% as described above) were plated in triplicate in 200 l of RPMI 1640 medium (supplemented with 10% FCS, 5 ϫ 10 Ϫ5 M 2-ME, 2 mM L-glutamine, and antibiotics) in 96-well plates.…”
