Natural resistance to ascorbic acid induced oxidative stress is mainly mediated by catalase activity in human cancer cells and catalase-silencing sensitizes to oxidative stress

Abstract: BackgroundAscorbic acid demonstrates a cytotoxic effect by generating hydrogen peroxide, a reactive oxygen species (ROS) involved in oxidative cell stress. A panel of eleven human cancer cell lines, glioblastoma and carcinoma, were exposed to serial dilutions of ascorbic acid (5-100 mmol/L). The purpose of this study was to analyse the impact of catalase, an important hydrogen peroxide-detoxifying enzyme, on the resistance of cancer cells to ascorbic acid mediated oxidative stress.MethodsEffective concentratio… Show more

“…5F). Both SOD2 and CAT are antioxidant enzymes, and are associated with regulation of cellular ROS and the acquired drug resistance in cancer (15,17,35). Consistent with microarray results, RT-PCR analyses showed that the expression of SOD2 and CAT transcript was increased in enriched HN-CICs than that in parental HNSCCs (Fig.…”

“…Availability of excessive ASC has been linked with an increase in the hydrogen peroxide formation that may consequently result in cell death [37]. This explains a slight increase in catalase activity as well as increased expression of catalase gene in Drosophila larvae from the ASC feeding regime.…”

“…It has been revealed that oxidative stress can be induced by tumor cells. Nrf2 is mutational and overexpressed in many tumor tissues and cell lines (27,28). Nrf2 transcription complex specifically recognizes ARE, which is located in the promoter region of all Nrf2-targeted genes and subsequently initiates the expression of a group of genes.…”
