Background and Objective: Diabetes foot ulcer (DFU) is among the worst devastating diabetic complications, frequently leading to amputation and death. Chrysin is a potent flavonoid, with various biological activities including antidiabetic action. However, its efficacy in the prevention of wound healing in DFU is not yet documented. Therefore this study was intended to evaluate the efficacy of chrysin in healing streptozocin-induced diabetic foot ulcers (STZ). Materials and Methods: Animals were selected into four groups (n = 10) and the diabetes was induced with STZ (55 mg kgG 1 ). The diabetic animals were made wounds and treated with two different doses (40 and 80 mg kgG 1 b.wt.) of Chrysin. The boyʼs weight, food intake, serum glucose, wound contraction rate, HbA1C levels, antioxidants and oxidative stress markers levels were measured. Further, the mRNA expression level of inflammatory markers and protein expression of growth factors were also evaluated. Results: Chrysin treatment increased the percentage of wound contraction rate, reduced the HbA1C level and increased the antioxidants and hydroxyproline level and decreased MDA and MOP activity to normal as compared to diabetic wound (DW) rats. Further, Chrysin administration attenuated the elevated mRNA levels of TNF-", IL-6 and NF-κB mRNA to normal. In addition, the protein level of VEGF and TGF-$1 were markedly improved by Chrysin, implicating its wound healing potential. Conclusion: This study demonstrated that the administration of Chrysin could prevent DFU with enhanced wound healing potential.

Human pancreatic α-amylase (HPA), situated at the apex of the starch digestion hierarchy, is an attractive therapeutic approach to precisely regulate blood glucose levels, thereby efficiently managing diabetes. Polyphenols offer a natural and multifaceted approach to moderate postprandial sugar spikes, with their slight modulation in carbohydrate digestion and potential secondary benefits, such as antioxidant and antiinflammatory effects. Taking into consideration the unfavorable side effects of currently available commercial medications, we aimed to study a library of polyphenols attributed to their remarkable antidiabetic properties and screened the most potent HPA inhibitor via a comprehensive in silico study encompassing molecular docking, molecular mechanics with generalized Born and surface area solvation (MM/GBSA) calculation, molecular dynamics (MD) simulation, density functional theory (DFT) study, and pharmacokinetic properties followed by an in vitro assay. Significant hydrogen bonding with the catalytic triad residues of HPA, prominent MM/ GBSA binding energy of −27.03 kcal/mol, and the stable nature of the protein−ligand complex with regard to 100 ns MD simulation screened quercetin as the best HPA inhibitor. Additionally, quercetin showed strong reactivity in the substrate-binding pocket of HPA and exhibited favorable pharmacokinetic properties with a considerable inhibitory concentration (IC 50 ) of 57.37 ± 0.9 μg/mL against α-amylase. This study holds prospects for HPA inhibition and suggests quercetin as an approach to therapy for diabetes; however, it is imperative to conduct further research.

Millions of people in developing nations rely on herbal traditional medicine for the treatment of ailments such as diabetes mellitus, stomach disorders and respiratory diseases. Xeroderris stuhlmannii (Taub.) Mendonca & E.P. Sousa is a medicinal plant used traditionally in Zimbabwe to treat diabetes mellitus and its complications. However, there is no scientific evidence to support its role as an antidiabetic medicinal plant. Here we hypothesized that Xeroderris stuhlmannii (Taub.) Mendonca & E.P. Sousa contain bioactive phytochemicals that can scavenge free radicals, and inhibit digestive enzymes that contribute to type 2 diabetes mellitus (T2DM). To test this hypothesis, we examined the free radical scavenging potential of crude extracts using the diphenyl-2-picrylhydrazyl assay in vitro. Furthermore, we carried out in vitro antidiabetic activity of crude extracts using chromogenic 3,5-dinitrosalicylic acid and p-nitrophenyl-alpha-D-glucopyranoside substrates on alpha-amylase and alpha-glucosidase. In addition, we used molecular docking approaches to screen for bioactive phytochemical compounds targeting the digestive enzymes. Our results showed that phytochemicals in Xeroderris stuhlmannii (Taub.) Mendonca & E.P. Sousa extracts scavenged free radicals with IC50 values ranging from 0.011-0.013 micrograms/mL. Further, the crude extracts significantly inhibited alpha-amylase and alpha-glucosidase with IC50 values of 12.9-21.1 micrograms/mL and 8.8-16.0 micrograms/mL, respectively. In silico molecular docking findings and pharmacokinetic predictions showed that myricetin is a novel inhibitor of the digestive enzymes that contributes to high blood glucose. Collectively, our findings suggest pharmacological targeting of digestive enzymes by Xeroderris stuhlmannii (Taub). Mendonca & E.P. Sousa crude extracts could lesion type 2 mellitus complications in humans.
