Natural Killer Cell Receptor Expression by Human First Trimester Decidual Granular Leukocytes and T‐Lymphocytes

Abstract: Like peripheral blood NK cells, decidual NK cells express the natural cytotoxicity receptors NKp30 and NKp46 but the significance of this will not become apparent until ligands for these molecules have been identified. Only a minority of decidual T cells express CD158, indicating that this is not a mechanism for inhibition of cytotoxicity mediated by all decidual T cells.

“…NKp30 mRNA and protein are known to be expressed by first trimester decidual uterine NK cells [11,12]. Data from our transcriptional profiling of human endometrium showed that NKp30 mRNA was significantly upregulated during the late-secretory stage of the menstrual cycle [1].…”

“…Given that NKp30 is expressed by both resting and NK cells and it has been shown to be expressed by decidual uNK cells [11,12], it was surprising to find no uNK immune reactivity in the stroma of late-secretory phase endometrium by IHC (even though there were many uNK cells present, Fig. 2N).…”

“…Previous studies of KIR expression in uNK cells used patients whose genotype was not known (5,6,26). In addition the Abs used were not able to discriminate between activating and inhibitory KIR.…”

“…In an immunogenetic study, we showed that specific combinations of two polymorphic gene systems, killer Ig-like receptors (KIR) in the mother and HLA-C in the fetus were associated with pre-eclampsia (4). In the decidua, KIR are expressed only by uNK cells and their cognate ligands include HLA-C molecules that are expressed by trophoblast (5)(6)(7)(8).…”
