Natural killer cell memory

Abstract: Natural killer (NK) cells are bone marrow–derived granular lymphocytes that have a key role in immune defense against viral and bacterial infections and malignancies. NK cells are traditionally defined as cells of the innate immune response because they lack RAG recombinase–dependent clonal antigen receptors. However, evidence suggests that specific subsets of mouse NK cells can nevertheless develop long-lived and highly specific memory to a variety of antigens. Here we review published evidence of NK cell–med… Show more

“…Of course, many other cell types are involved in the regulation of antitumor immunity, and the effects of TriMix on these different cells is not yet known. For instance, it might be possible that TriMix transfected DCs could stimulate natural killer (NK) cells, which act synergistically with CTLs and were recently reported to mount antigen-specific responses that can lead to long-term memory [38][39][40][41].…”

“…In 2006, the observation that mice lacking T cells and B cells could develop CHS responses to various distinct haptens introduced the concept that NK cells can mediate antigen-specific memory responses 12,13 . Several lines of evidence support the notion that NK cells can contribute to immunological memory.…”

“…Despite these known innate immune cell functions, accumulating evidence in both mice and humans demonstrates that NK cells are educated and selected during development, possess receptors with antigen specificity, undergo clonal expansion during infection and can generate long-lived memory cells. 33,34 After over 30 years of researching NK cells, evidence supports that they play critical roles in the early control of viral infection, in hematopoietic stem cell (HSC) transplantation (improved grafting, graft-vs.-host disease and graft-vs.-tumor), in tumor immunosurveillance and in reproduction (uterine spiral artery remodeling). The roles of NK cells in controlling organ transplantation, parasitic and HIV infections, autoimmunity and asthma have also been suggested, but remain to be explored further.…”
