Natural Field Spread of Mild and Severe Isolates of Citrus Tristeza Virus in Florida

Abstract: The effectiveness of five different aphid control regimes in delaying movement of mild and severe isolates of citrus tristeza virus (CTV) into a CTV-free sweet orange scion on sour orange rootstock block was monitored annually for 5 years, using severe isolate-specific and isolate-nonspecific monoclonal antibodies. The average percentage of trees infected with a severe isolate of CTV was 32, 32, 20, 25, and 28 for trees treated with Temik, Temik + Meta-Systox R (MSR), MSR, stylet oil, or no aphid control, resp… Show more

“…The epidemiological studies on the use of oils as a control strategy has been mainly conducted with non-persistently aphid-transmitted viruses (Perring et al, 1999), including under nursery blocks conditions (Vidal et al, 2010). Furthermore, the use of mineral oil treatments has also been successfully tested against semi-persistent and persistent viruses (Allen et al, 1993;Perring et al, 1999;Asjes & Blom-Barnhoorn, 2001), including CTV (Powell et al, 1997).…”

“…Routine detection of the virus is mainly performed by serological methods [enzyme‐linked immunosorbent assay (ELISA) and tissue‐print ELISA] based on polyclonal or specific monoclonal antibodies (Mathews et al., 1997; Powell et al., 1997; Hughes and Gottwald, 1998; Cambra et al., 2000a,b), although nucleic acid‐based methods have been developed, such as polymerase chain reaction (PCR)‐based assays performed on immunocapture (Nolasco et al., 1993) or print or squash capture (Olmos et al., 1996; Cambra et al., 2000b), nested‐PCR in a single closed tube (Olmos et al., 1999) and a co‐operational PCR system (Olmos et al., 2002). The best option depends on the particular purpose.…”
