Natural cytotoxicity in man: activity of lymph node and tumor‐infiltrating lymphocytes

Abstract: Lymphocytes from blood, lymph node and tumor have been tested for cytotoxicity against the K562 cell line which is known to be highly sensitive to lysis by spontaneously reactive cells. Cytotoxicity was found in all 13 samples from healthy donors and in 17/32 cancer patients. By contrast, activity was determined in only 1/18 lymph node and 1/14 preparations of tumor-infiltrating lymphocytes. Lymph node cells were similarly nonreactive against 3 other cell lines known to be sensitive to natural cytotoxicity. St… Show more

“…Even so, there are likely to be few: since up to 60% of HNK1 + cells are also OKM1+ (Abo et al, 1982a) it would appear that most of the OKM1 + cells in the tumours are macrophages. This conclusion accords with functional studies conducted with lymphoid cells recovered from freshly disaggregated neoplasms wherein breast, in common with other solid tumours has generally disclosed low or non-existent levels of NK activity, (Vose et al, 1977;Totterman et al, 1978;Moore & Vose, 1981;Eremin et al, 1981;Introna et al, 1983). The recent availability of McAbs with greater selectivity for NK cells should allow further examination of this question (Perussia et al, 1983a, b …”

“…In general, NK-cell function was markedly depressed in human tumours (Vose et al, 1977a;Totterman et al, 1980;Mantovani et al, 1980a;Haskill et al, 1982b). Specifically cytotoxic T cells have only been detected in a few instances (Vose et al, 1977b;Werkmeister et al, 1979) suggesting that effector-cell function may be difficult to maintain in situ.…”

“…Enriched preparations of ascitesassociated lymphocytes were obtained by stepwise density and velocity separations similar to those used by Vose et al (1977) followed by repeated adherence steps or discontinuous Ficoll-Hypaque gradients. The percentage of lymphoid cells forming E RFC (39%) averaged less than that in either blood from patients or normal blood.…”
