Natural Antimicrobial Peptides as Inspiration for Design of a New Generation Antifungal Compounds

Abstract: Invasive fungal infections are associated with high mortality rates, despite appropriate antifungal therapy. Limited therapeutic options, resistance development and the high mortality of invasive fungal infections brought about more concern triggering the search for new compounds capable of interfering with fungal viability and virulence. In this context, peptides gained attention as promising candidates for the antimycotics development. Variety of structural and functional characteristics identified for vario… Show more

“…Najnowsze doniesienia literaturowe wskazują, że właściwe wykorzystanie katelicydyn lub odpowiednia stymulacja ich endogennej biosyntezy w organizmie, może przyczynić się do zniwelowania zagrożenia ze strony chorobotwórczych bakterii [5,22,23,26,27] oraz grzybów [28]. Infekcje C. albicans są istotnym problemem epidemiologicznym, a około połowa populacji to stali nosiciele tego grzyba.…”

“…Antimicrobial peptides (AMPs) have been identified in virtually all organisms and have diverse structures and functions, such as antimicrobial and immunomodulatory activities [4][5][6][7][8]. Because AMPs exhibit broad-spectrum activity against microorganisms and insusceptibility to conventional drug resistance mechanisms, AMPs are promising candidates for the development of new antifungal drugs [9][10][11][12][13].…”

“…Existing within nature as mediators of the innate immune response, they are particularly promising given they possess multiple microbial targets, thereby reducing chance of resistance development. Microbial targets include fungal membranes and molecules involved within intracellular biomolecular processes such as protein replication and DNA synthesis [17]. Key to antimicrobial efficacy and selectivity is the hydrophobic: charge balance of the peptide motif, which ultimately defines membrane interactions and surfactant-like activity [18].…”
