We previously isolated an analog to chlorophyll-related compounds, pheophytin a, from the marine brown alga Sargassum fulvellum and demonstrated that it is a neurodifferentiation compound. In the current study, we investigated the effects of the pheophytin a analog vitamin B 12 on PC12 cell differentiation. In the presence of a low level of nerve growth factor (10 ng ml -1 ), vitamin B 12 demonstrated neurite outgrowthpromoting activity in PC12 cells. The effect was dose-dependent in the range of 6-100 lM. In the absence of nerve growth factor, vitamin B 12 did not promote differentiation. To investigate the mechanism for this effect, we conducted differentiation assays and western blot analysis with signal transduction inhibitors and found that vitamin B 12 did not promote PC12 cell differentiation in the presence of K252a or U0126 inhibitors. These results suggest that vitamin B 12 stimulates PC12 cell differentiation through enhancement of the mitogen-activated protein kinase signal transduction pathway, which is also induced by nerve growth factor. Thus, vitamin B 12 may be a good candidate for treatment of neurodegenerative diseases such as Alzheimer's disease.

ObjectivesNJ is being used to treat inflammatory diseases in Korea. In this study, we attempted to evaluate the effects of NJ on atopic dermatitis (AD)-like lesions and mast cell-mediated allergy inflammation in vivo and in vitro. Methods and ResultsWe investigated to ascertain the pharmacological effects of NJ on 2,4-dinitrofluorobenzene (DNFB)-induced allergic reactions under in vivo conditions. Additionally, to find possible explanations for the anti-inflammatory mechanisms of NJ, we evaluated the regulatory effects of NJ on the level of inflammatory mediators in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated human mast cells (HMC-1). ConclusionsNJ inhibited the production of the inflammatory cytokines (IgE, IL-6, IL-8 and TNF-α) significantly in vivo and in vitro.

In the course of our investigations of neuritogenically active substances from medicinal plants we have devoted our attention to the occurrence of natural products having new skeletons. We isolated a new dimeric dihydrochalcone, littorachalcone (1), which showed a considerable activity at enhancing nerve growth factor-mediated neurite outgrowth from PC12D cells, from Verbena littoralis H. B. K. (Verbenaceae) by a series of bioassay-directed chromatographic separations. Compounds that possess properties which enhance the ability of a nerve growth factor to stimulate neurite outgrowths from PC12D cells may be useful in the treatment of neurological disorders, such as Parkinson's disease (PD), Alzheimer's disease (AD), Huntington's disease (HD), amyotrophic lateral sclerosis (ALS), and human immunodeficiency virus associated dementia (HAD). 1-7)The EtOAc soluble materials of MeOH extracts of the aerial parts of V. littoralis collected in Paraguay were repeatedly subjected to silica gel columns (EtOAc/MeOH) and Sephadex LH-20 column (MeOH) followed by reversedphase semi-preparative HPLC on a YMC-pack ODS-AM324 column (78% MeCN in H 2 O) to yield littorachalcone (1), together with two known flavonoids, 4Ј-hydroxywogonin (2) and 8,3Ј-dimethoxy-5,7,4Ј-trihydroxyflavone (3).Littorachalcone (1) showed a molecular ion peak at m/z 514 in the electron impact (EI)-MS spectrum, and its molecular formula was determined to be C ) moieties. The structural characterization of 1 was performed by analyzing its EI-MS and one dimensional (1D) and 2D NMR spectroscopic data (see Table 1, Figs. 1-3). The 1 H-NMR spectrum disclosed the presence of 13 signals for aromatic hydrogen atoms, eight signals for methylene hydrogens, and two hydroxyl groups. Analysis of the 13 C-NMR, distortionless enhancement by polarization transfer (DEPT), and 1 H-detected heteronuclear multiple quantum coherence (HMQC) spectra of 1 indicated the presence of 24 aromatic carbons, four sp 3 methylene carbons, and two carbonyls. These features of the 1 H-and 13 C-NMR spectra of 1 were similar to those of verbenachalcone. A new dihydrochalcone dimer, 2,4,3؆,2ٟ,4ٟ-pentahydroxy-4-O-4؆-tetrahydrobichalcone, given the name littorachalcone, was isolated from the aerial parts of Verbena littoralis H. B. K. along with two known flavonoids 4-hydroxywogonin and 8,3-dimethoxy-5,7,4-trihydroxyflavone. Littorachalcone caused a significant enhancement of nerve growth factor-mediated neurite outgrowth from PC12D cells.
