Narciclasine, a potential allelochemical, affects subcellular trafficking of auxin transporter proteins and actin cytoskeleton dynamics in Arabidopsis roots

Abstract: The present study documented the action of a potential allelochemical, narciclasine, on auxin transport in Arabidopsis by mainly affecting subcellular trafficking of PIN and AUX1 proteins and through interfering actin cytoskeletal organization. Narciclasine (NCS), an Amaryllidaceae alkaloid isolated from Narcissus tazetta bulbs, has potential allelopathic activity and affects auxin transport. However, little is known about the cellular mechanism of this inhibitory effect of NCS on auxin transport. The present … Show more

“…Compared to other AMPK activators, narciclasine stimulates the kinase in vivo at low doses and shows efficacy in cancer, metabolic and pain models at doses that coincide with AMPK activation kinetics (Van Goietsenoven et al, 2010, Julien et al, 2017a, Julien et al, 2017b, Inyang et al, 2019b). Mechanistically, narciclasine increases the adenosine diphosphate (ADP)/ATP ratio in myotubes, which is a potential mechanism through which the compound causes AMPK activation (Hu et al, 2015, Julien et al, 2017a, Julien et al, 2017b). The mechanism through which narciclasine activates AMPK in sensory neurons is not known, but our previous work demonstrates that the drug stimulates AMPK in dorsal root ganglion neurons in vitro and in vivo (Inyang et al, 2019b).…”

“…Besides, also during lateral root organogenesis, cytokinin enhances the PIN1 depletion from cell membrane to affect PIN1 polarization [ 53 55 ]. Furthermore, it is reported that the localization of PIN proteins is affected by diffusible molecules such as jasmonate and narciclasine [ 56 , 57 ]. It is not yet clear whether these molecules are involved in the phyllotaxis pattern or not.…”
