Nanosuspensions as delivery system for gambogenic acid: characterization and in vitro/in vivo evaluation

Abstract: Nanosuspensions (NS) can enhance the saturation solubility and dissolution velocity of poorly soluble drugs. PEG as a non-ionic surfactant plays an important role in surface modification of nanoparticles for prolonging in vivo circulation. In this study, anti-solvent precipitation method was introduced to prepare gambogenic acid nanosuspensions (GNA-NS) with PVPK30 and PEG2000 as stabilizers to settle the disadvantages of GNA. The obtained nanoparticles were spherical with a mean particle size of 183.7 nm and … Show more

“…All measurements were conducted in triplicate. [41][42][43] The surface morphology of HA/ Zein-DOX NPs was observed by transmission electron microscope (TEM, Japan electronics corporation, JEM-1400, Tokyo, Japan). HA/Zein-DOX NPs were gold coated (300 A) with an Edwards S150B sputter coater to improve electrical conductivity and observed under the SEM.…”

“…In another study, the plant bioactive compound gambogenic acid nanosuspensions were developed using the solvent precipitation method with the particle size of about 183 nm with higher anticancer efficacy than gambogenic acid. 120 However, microfluidization-based nanosuspension will be an alternative approach in the delivery of many plant bioactive compound-based nanosuspensions with higher efficacy in the bioavailability of those compounds against various chronic diseases.…”

“…Despite the promising pharmacological activities of GNA, its disadvantages such as low water solubility, low oral bioavailability, short half‐life and strong vascular stimulation severely limited its clinical application . In order to improve the bioavailability and therapeutic efficacy, many efforts have been done, such as GNA‐loaded cubosomes, GNA‐loaded solid lipid nanoparticles, PEGylated non‐ionic surfactant vesicles of GNA, GNA nanosuspensions and long circulation nanostructured lipid carriers of GNA were successfully prepared in previous researches of our group …”
