This work was focused on identification and evaluation of process parameters of modified nanoprecipitation method, for fabrication of lomustine nanoparticles, with the aim of reducing cancer cell viability at low concentration of lomustine. The parameters controlling particle size, mostly in nanosize, were solvent/nonsolvent composition and emulsification speed of homogenizer along with aqueous phase volume. This controlled particle size is below 250 nm. The stabilizer concentration controlled particle size is within 68 nm ± 0.89 to 137 ± 0.94 nm with PDI 0.06 ± 0.008 to 0.25 ± 0.001. But, the stabilizer addition mode showed more uniform size distribution with PDI 0.085 ± 0.004. Entrapment efficiency was maintained well above 47 ± 0.23%. The drug release pattern was monophasic with controlled release over 24 hrs. In the method used, drug content was affected by ratio of polymer to drug to organic solvent, as well as homogenization speed and time. Percentage viable cells of L132 human lung cancer cell line remained, were only 5% at 100 µg/ml lomustine equivalent PLA nanoparticles. ethanol as a cosolvent, homogenization speed and time were evaluated affecting the particle size, encapsulation efficiency and in vitro drug release.

Novel gradient copolymers of hydrophilic 1-vinylimidazol and hydrophobic methacrylic derivative of ibuprofen prepared by free radical polymerization are described. The heterogeneous distribution of monomeric units along the polymeric chains leads to a gradient distribution of the hydrophobic and hydrophilic sequences responsible of nanoparticles formation through a self-assembling mechanism, capable of tune the water permeation due to the ionizable imidazole moieties and their gradient profile along the macromolecules, exhibiting pH and composition dependent effect in terms of diameter, zeta potential, acid-base buffering, ibuprofen release and chelating capacities, responsible of matrix metalloproteinase dysfunction showing anti-inflammatory activity in a nitric oxide inhibition assay.

Antibacterial amphiphiles based on ε-polylysine: synthesis, mechanism of action, and cytotoxicity

21The development of antibacterial materials is recently more and more important and 22 urgent due to the emergence of antibiotic-resistant bacteria. To address the problem, 23 series of new amphiphlies based on ε-polylysine (ε-PL) were synthesized by 24 alkylation reaction with alkyl bromide, and concurrently their antibacterial activity 25 and cytotoxicity were evaluated. The amphiphlies with a large concentration of 26 positive charges and lipid chain promoted their adsorption to bacterial membranes 27 through electrostatic interaction and hydrophobic interaction, subsequently killed both 28 gram-positive (S. aureus and B. amyloliquefaciens) and gram-negative (E. coli and P. 29 aeruginosa) bacteria. Morphology observed using SEM shows that these derivatives 30 could cause leakage of intracellular contents. Analysis of the antimicrobial 31 mechanism displays that these derivatives against the bacteria started with disruption 32 of the bacterial membrane, which caused the leakage of cytoplasm, and killed the 33 bacteria. Among the amphiphlies, ε-PL-g-butyl2 presented the most effective 34antibacterial activity and its minimum inhibitory concentration as low as 3.9 µg/mL. 35 Importantly, the effective antibacterial concentration of ε-PL-g-butyl2 displayed no 36 cytotoxicity against human cells. This work not only highlights the great promise of 37 using ε-PL-g-butyl2 as a highly effective antibacterial agent but also provides the 38 important tool for understanding the interactions between the microorganisms and 39 amphiphlies-based ε-PL. 40
