Nanoparticles for Diagnosis and Target Therapy in Pediatric Brain Cancers

Abstract: Pediatric brain tumors represent the most common types of childhood cancer and novel diagnostic and therapeutic solutions are urgently needed. The gold standard treatment option for brain cancers in children, as in adults, is tumor resection followed by radio- and chemotherapy, but with discouraging therapeutic results. In particular, the last two treatments are often associated to significant neurotoxicity in the developing brain of a child, with resulting disabilities such as cognitive problems, neuroendocri… Show more

“…In this context, targeted therapy is an attractive therapeutic approach. MEK inhibitors and BRAF V600E inhibitors have already demonstrated efficacy and are being compared to standard therapy in trials of treatment-naïve patients with LGG [30,31]. However, because these new targeted therapies frequently target genes and pathways that are critical in normal brain development, the acute and subacute long-term sequelae of molecular-targeted therapies need to be carefully monitored in children.…”

“…Furthermore, PLGA NPs conjugated with an anti-epidermal growth factor receptor (EGFR) monoclonal antibody and loaded with curcumin achieved a reduction in the growth of EGFR-expressing GBM cells at lower concentrations than those required for free curcumin or unmodified curcumin-loaded PLGA NPs to achieve this effect [ 75 ]. Lipid-based and polymeric NPs are also being explored for the delivery of chemotherapeutic agents in paediatric cancers (for review see Guido et al, 2022 [ 76 ]).…”

“…Imaging contrast parameters-based nanotechnology are being updated every day and are specifically presented an ability for detection the tumors in vivo compared with conventional scanning devices [14][15][16] . Furthermore, the platforms of nanoscale imaging have novel patterns with a lower health risk and a higher penetration 17 .…”
