Nanoliposomes from Agro-Resources as Promising Delivery Systems for Chondrocytes

Abstract: Investigations in cartilage biology have been hampered by the limited capacity of chondrocytes, especially in rats and humans, to be efficiently transfected. Liposomes are a promising delivery system due to their lipid bilayer structure similar to a biological membrane. Here we used natural rapeseed lecithin, which contains a high level of mono- and poly-unsaturated fatty acids, to evaluate the cytocompatibility of these phospholipids as future potential carriers of biomolecules in joint regenerative medicine.… Show more

“…All these features allow liposomal nanoformulations to improve the drug therapeutic index and decrease its side effects [20]. Made of phospholipids, amphiphilic molecules commonly found in cells membrane and essential for its structure [21][22][23][24], liposomes can encapsulate and protect hydrophobic drugs in its membrane before fusion with the targeted cell membranes, and thus allowing the drug release at sufficient concentration [25]. Moreover, the surface of liposomes can also be functionalized to further increase its targeting efficiency [26,27].…”

“…Liposomes have been studied and investigated for more than five decades and have become a well-established drug delivery vesicle, resulting in the marketing authorization of many clinically approved liposome-based products to treat different diseases [ 1 ]. Liposomes’ resemblance to biomembranes enables superior biocompatibility and safety over other polymeric and metal-based nanoparticles, as well as the ability to deliver lipid-soluble and water-soluble molecules at the same time [ 2 , 3 ]. However, liposomes require surface modification with ligands to acquire smart targeting capabilities.…”

“…Chondrocytes are surrounded by an extracellular matrix (ECM) they synthetize. Articular cartilage viscoelastic properties provide wear-resistant surfaces to the joint by improving the distribution of mechanical loads and moderating the friction between its surfaces (Sanchez-Adams et al, 2014;Lindahl, 2015;Bianchi et al, 2020;Masson and Krawetz, 2020). When injured or degenerating, articular cartilage is not able to restore its original organization because it has a very weak regenerative capacity.…”
